生长素类物质对甘薯茎尖分生组织培养成苗的影响 预览 被引量:9

Effects of auxins on plantlets regeneration from subcultured tip meristem in sweet potato

摘要 以豫薯4号和豫薯12号为试材,研究了生长素类物质(IBA和NAA)、温度和光照时间对转移后甘薯茎尖分生组织培养成苗的影响.结果表明,适宜甘薯茎尖分生组织培养成苗的温度为28℃,光照时间以不低于14h@d-1为好,培养基中加入0.1~0.5mg/L的IBA(或NAA)有利于甘薯茎尖分生组织培养成苗,IBA的效果优于NAA.与不加任何生长素类物质的MS培养基相比,添加IBA 0.5mg/L的MS培养基可使甘薯茎尖分生组织培养成苗率提高9.9%~15.4%,平均成苗天数缩短3.3~8.5d.IBA(或NAA)浓度过高时(达到1.0mg/L),甘薯茎尖外植体形成大量的愈伤组织,导致成苗率降低,平均成苗天数延长. Yushu 4 and Yushu 12 were used to study the effects of auxins(IBA and NAA),temperature and illumination time on plantlets regeneration from subcultured tip meristem in sweet potato.The results showed that the suitable temperature was 28℃,illumination time was more than 14h·d-1 .01-05 mg/L IBA(or NAA) into MS medium was beneficial to plantlets regeneration from subcultured tip meristem in sweet potato.The effect of IBA was superior to NAA.In comparison with MS medium without any auxins.the MS medium supplemented with IBA 05mg/L could increase tip meristem culture plantlet ratio by 99%-154% and reduce mean time of plantlet by 33-85d.When concentration of IBA(or NAA) was high(to 10 mg/L),tip meristem explant of sweet potato pruduced much callus,plantlet ratio from subcultured tip meristem in sweet potato decreased and the average plantlet time was delayed.
