细虫草无性型——羽束梗孢胞内多糖的研究

摘要 从新疆细虫草Ophiocordyceps grsacilis的子实体分离出无性型——羽束梗孢Paraisaria dubia,为细虫草开发应用奠定基础,增加虫草新资源。经液体发酵培养10 d产生白色小菌球状的菌丝体和清澈透明发酵液,菌丝体经热水提取、乙醇沉淀得胞内粗多糖,再经乙醇脱蛋白得脱蛋白多糖,最后经Sepharose 4B柱色谱纯化得胞内纯多糖。对胞内多糖进行紫外光谱,红外光谱,核磁共振氢谱、碳谱,气相色谱,高效液相等分析。结果表明羽束梗孢胞内多糖由葡萄糖、半乳糖、甘露糖组成,其摩尔比为25. 54∶2∶1,含有吡喃糖苷,为β糖苷键构型,初步获得多糖连接方式为β(1→2)(1→4)(1→6)连接。该实验为羽束梗孢的开发应用提供理论依据。 In order to provide a foundation for the development and application of Ophiocordyceps gracilis and increase the new resources of cordyceps,an asexual Paraisaria dubia was isolated from an O.gracilis fruit body.After 10 days of liquid fermentation,white globular mycelium and clear transparent fermentation were produced.The mycelium was extracted by hot water and precipitated with ethanol to obtain intracellular crude polysaccharide.The protein was deproteinized to obtain deproteinized polysaccharide.The intracellular pure polysaccharide was purified by Sepharose 4 B column chromatography and were analyzed by UV,IR,1 H-NMR,and13 CNMR data,as well as GC and HPLC.The results showed that the intracellular polysaccharide of P.dubia was composed of glucose,galactose and mannose with a molar ratio of 25.54∶2∶1.It was aβ-configuration glycosylic bond,containing pyranoside.The initial connection of polysaccharide wasβ(1→2)(1→4)(1→6)connection.This experiment provides a theoretical basis for the development and application of P.dubia.
