Thrips-mediated impacts from transgenic rice expressing Cry1Ab on ecological fitness of non-target predator Orius tantilus(Hemiptera:Anthocoridae) 预览

Various rice lines have been genetically modified with genes from Bacillus thuringiensis(Bt) to continuously produce Bt insecticidal proteins against lepidopteran larvae. The Bt insecticidal protein constantly expresses in the plants to create an opportunity for non-target herbivores to acquire and convey the protein to their predators or parasitoids across trophic levels. This paper evaluates the effects of Bt rice(namely, Kemindao 1(KMD1) and Kemindao 2(KMD2)) expressing Cry1 Ab as compared to its non-Bt control line, Xiushui 11 on non-target predator Orius tantilus(a generalist predatory anthocorid of thrips) under laboratory and field conditions. To measure several biological parameters such as total nymphal duration and fecundity of this bug, it was reared on thrips and pollens of KMD1 and KMD2 as compared to their control under laboratory conditions. By comparison with the control, Bt rice did not significantly affect main life-history characteristics(total nymphal duration, female adult longevity, oviposition period and fecundity) of this anthocorid preying on Bt rice-fed thrips along with Bt rice pollens, except that the fecundity of this predator for KMD1 was distinctly lower as compared with KMD2 or the control. Enzyme-linked immunosorbent assay(ELISA) results showed that no Cry1 Ab protein was detected in this predator fed on thrips or rice pollen from Bt rice but was in Bt rice pollens. With the beat plate, plastic bag and color trap sampling methods, two-year field monitoring of O. tantilus abundance demonstrated that Bt rice had no significant detrimental effects on the population dynamics and seasonal average densities of this predatory anthocorid as compared with the control. Thus, it is suggested that growing our tested Bt rice(KMD1 and KMD2) producing Cry1 Ab will pose a negligible risk to the anthocorid, O. tantilus. 展开更多

Safe and efficient gene transfer systems are the basis of gene therapy applications. Non-integrating lentiviral(NIL) vectors are among the most promising candidates for gene transfer tools, because they exhibit high transfer efficiency in both dividing and non-dividing cells and do not present a risk of insertional mutagenesis. However, non-integrating lentiviral vectors cannot introduce stable exogenous gene expression to dividing cells, thereby limiting their application. Here, we report the design of a non-integrating lentiviral vector that contains the minimal scaffold/matrix attachment region(S/MAR) sequence(SNIL), and this SNIL vector is able to retain episomal transgene expression in dividing cells. Using SNIL vectors, we detected the expression of the eGFP gene for 61 days in SNIL-transduced stable CHO cells, either with selection or not. In the NIL group without the S/MAR sequence, however, the transduced cells died under selection for the transient expression of NIL vectors. Furthermore,Southern blot assays demonstrated that the SNIL vectors were retained extrachromosomally in the CHO cells. In conclusion,the minimal S/MAR sequence retained the non-integrating lentiviral vectors in dividing cells, which indicates that SNIL vectors have the potential for use as a gene transfer tool. 展开更多

Pigs do not secret β-glucanase to degrade the β-glucan in their feed. The indigestible β-glucan not only blocks the release of nutrients from the grain cell wall, but also increases the digesta viscosity in the gastrointestinal tract of pigs. Therefore, dietary β-glucan significantly inhibits nutrient digestion and absorption in pigs. Transgenic expression of β-glucanase in the digestive tract of pigs may offer a solution to solve this problem. In the current study, four artificial codon-optimized β-glucanases genes was prepared and expressed in porcine cells. Only p Bg A and p Egx showed high activity in transfected pig kidney cells. To improve the p H range and p H stability of β-glucanase, the two β-glucanases, p Bg A and p Egx, were co-expressed in pig kidney cells and salivary gland cells by Linker A3 or 2A peptide. The resulting dual enzymes of p Bg A3 p Eg and p Bg2 Ap Eg showed significantly enlarged p H range and significantly increased p H stability, as compared to parental enzymes. These results provide useful data for future study on increasing the feed digestibility of pigs by transgenic expression of β-glucanase in their salivary glands. 展开更多

为研究山茶花CjAPL1基因对于重瓣花形成的作用,构建了pCAMBIA1300-CjAPL1正义植物表达载体,酶切结果显示,CjAPL1基因正向插入pCAMBIA1300的启动子和终止子之间,表明CjAPL1植物表达载体构建成功。将pCAMBIA1300-CjAPL1采用农杆菌介导的... 为研究山茶花CjAPL1基因对于重瓣花形成的作用,构建了pCAMBIA1300-CjAPL1正义植物表达载体,酶切结果显示,CjAPL1基因正向插入pCAMBIA1300的启动子和终止子之间,表明CjAPL1植物表达载体构建成功。将pCAMBIA1300-CjAPL1采用农杆菌介导的花序浸染法转化拟南芥,获得转基因阳性植株65株。随机挑选表型变异明显的3株进行PCR扩增都得到了目的条带,Southern杂交鉴定进一步确认为转基因阳性植株。同时荧光定量检测到在转基因植株中CjAPL1基因表现出较对照显著提高6~25倍的表达量。转基因阳性植株表型变异明显,花柱和雄蕊数相比野生型各增加1枚和1~4枚,萼片边缘发育成白色的瓣化状,表明在拟南芥中过量表达CjAPL1基因可以引起花器官形态的变异和数量的变化,因此该基因在花器官形成和重瓣花发育中具有显著的调控功能。 展开更多

OBJECTIVE:In the present study,we systemically evaluated the ability of two bioactive compounds from traditional Chinese medicine,celastrol and pristimerin,to enhance recombinant adeno-associated virus(rAAV) serotype vector-mediated transgene expression both in human cell lines in vitro,and in murine hepatocytes in vivo.METHODS:Human cell lines were infected with rAAV vectors with either mock treatment or treatment with celastrol or pristimerin.The transgene expression,percentage of nuclear translocated viral genomes and the ubiquitination of intracellular proteins were investigated post-treatment.In addition,nonobese diabetic/severe combined immunodeficient gamma(NSG) mice were tail vain-injected with rAAV vectors and co-administered with either dimethyl sulfoxide,celastrol,pristimerin or a positive control,bortezomib.The transgene expression in liver was detected and compared over time.RESULTS:We observed that treatment with pristimerin,at as low as 1 umol/L concentration,significantly enhanced rAAV2 vector-mediated transgene expression in vitro,and intraperitoneal coadministration with pristimerin at 4 mg/(kgd) for 3 d dramatically facilitated viral transduction in murine hepatocytes in vivo.The transduction efficiency of the tyrosine-mutant rAAV2 vectors as well as that of rAAV8 vectors carrying oversized transgene cassette was also augmented significantly by pristimerin.The underlying molecular mechanisms by which pristimerin mediated the observed increase in the transduction efficiency of rAAV vectors include both inhibition of proteasomal degradation of the intracellular proteins and enhanced nuclear translocation of the vector genomes.CONCLUSION:These studies suggest the potential beneficial use of pristimerin and pristimerincontaining herb extract in future liver-targeted gene therapy with rAAV vectors. 展开更多

The harpin protein Hpa1 produced by the rice bacterial blight pathogen promotes plant growth and induces plant resistance to pathogens and insect pests. The region of 10-42 residues(Hpa110-42) in the Hpa1 sequence is critical as the isolated Hpa110-42 fragment is 1.3-7.5-fold more effective than the full length in inducing plant growth and resistance. Here we report that transgenic expression of Hpa110-42 in wheat induces resistance to English grain aphid, a dominant species of wheat aphids. Hpa110-42-induced resistance is effective to inhibit the aphid behavior in plant preference at the initial colonization stage and repress aphid performances in the reproduction, nymph growth, and instar development on transgenic plants. The resistance characters are correlated with enhanced expression of defense-regulatory genes(EIN2, PP2-A, and GSL10) and consistent with induced expression of defense response genes(Hel, PDF1.2, PR-1b, and PR-2b). As a result, aphid infestations are alleviated in transgenic plants. The level of Hpa110-42-induced resistance in regard to repression of aphid infestations is equivalent to the effect of chemical control provided by an insecticide. These results suggested that the defensive role of Hpa110-42 can be integrated into breeding germplasm of the agriculturally significant crop with a great potential of the agricultural application. 展开更多

In this study, recombinant baculovirus carrying the microdystrophin and β-catenin genes was used to infect adipose-derived stem cells from a dystrophin-utrophin double knock-out mouse. Results showed that, after baculovirus transgene infection, microdystrophin and β-catenin genes were effectively expressed in adipose-derived stem cells from the dystrophin-utrophin double knock-out mouse. Furthermore, this transgenic expression promoted adipose-derived stem cell differentiation into muscle cells, but inhibited adipogenic differentiation. In addition, protein expression related to the microdystrophin and Wnt/β-catenin signaling pathway was upregulated. Our experimental findings indicate that baculovirus can successfully deliver the microdystrophin and β-catenin genes into adipose-derived stem cells, and the microdystrophin and Wnt/β-catenin signaling pathway plays an important role in myogenesis of adipose-derived stem cells in the dystrophin-utrophin double knock-out mouse. 展开更多
