PKU-TA00001中发现了五个色胺类天然产物(1-5),其中化合物1和2是新化合物,化合物4是新天然产物,化合物3和4的核磁共振波谱数据首次被提供。生物活性筛选结果显示,所有化合物对几株革兰氏阳性菌和阴性菌的最低抑制浓度(MIC)均在50μM以上,对人肿瘤细胞株A549, HCT-8和MCF-7在100μM下未显示细胞毒活性。化合物1-4的发现扩展了色胺类天然产物的结构多样性,并为将来的生物合成和更多生物活性研究打下了基础。 展开更多

Samples of peels and soils from cassava waste sites were collected from cassava processing factory in Oyo town, Oyo State and also a dumpsite in Alaya village, Aiyedire local government, Osun State, Nigeria. Bacterial isolation was carried out using the pour plate method to obtain amylolytic bacterial species capable of hydrolyzing starch. Fourteen bacterial isolates that were most responsive to starch hydrolysis were selected from both sampling sites for molecular investigations. DNA of the isolates was extracted and subjected to a cocktail mix and condition for PCR which was purified using two universal primers and afterwards the PCR product was used for the polymorphism through electrophoresis. Bacterial isolates were identified based on their genetic sequences and results showed Bacillus subtilis (21.44%) to be the most frequently occurring specie. There was the prevalence of two similar strains Bacillus subtilis MML2483 and B. subtilis MML2411 which was isolated from the different sampling sites. Some other bacterial strains included Bacillus olivae BRB18, B. licheniformis HT-26-B1, B. cereus H17, B. safensis MS40, B. pumilus 07 展开更多

Protective Effect of Bacillus subtilis Peptidoglycan (PG) on β-conglycinin-induced Intestinal Epithelial Cells Damage of Juvenile Carp (Cyprinus carpio) 预览

[Objective] The paper was to investigate the protective effects of different concentrations of Bacillus subtilis peptidoglycan(PG) on β-conglycinin-induced inflammatory injury in intestinal epithelial cells of juvenile carp(Cyprinus carpio).[Method] In 24-cell microplates, the intestinal epithelial cells(IECs) of juvenile carp were primarily cultured for 72 h at 26°C and 6% CO2, and then the IECs were randomly divided into6 groups with 4 replicates per group. One of the six groups was set as negative control group, and the other groups were all supplemented with 1.0 mg/mL β-conglycinin in culture medium to establish inflammatory injury. At 24 h post induction, the culture media were changed into B. subtilis PG culture media with the concentrations of 0(positive control group), 0.15, 0.30, 0.45 and 0.60 mg/mL, respectively. The samples were collected to measure the antioxidant and anti-inflammatory indices at 12, 24 and 36 h post culture.[Result]β-conglycinin exposure significantly decreased the activity of ASA, AHR, SOD, CAT, GPx, and increased the PC content and the mRNA expression of inflammatory cytokines(IL-1β, IL-8, TNF-α1,IL-10 and TGF-β). At 12, 24 and 36 h post PG treatment, the activities of ASA, AHR, SOD, CAT, GPx and the content of PC in cells decreased in a dose-dependent manner;the mRNA levels of IL-1β, IL-8 and TNF-α1 were down-regulated and those of IL-10 and TGF-β were up-regulated.[Conclusion] Different concentrations of B. subtilis PG could protect IECs oxidative damage induced by β-conglycinin and improve the antioxidant capacity of IECs. High concentration of PG could improve the anti-inflammatory ability of IECs by inhibiting inflammatory factors and promoting the gene expression of anti-inflammatory cytokines. 展开更多

Traditional Chinese medicine Pseudostellaria heterophylla fibrous roots have been widely studied and applied in non-antibiotic clinical breeding of livestock and poultry. In vitro probiotic effect of P. heterophylla fibrous roots extract on feed probiotics Bacillus subtilis BD-K010 was studied, and the feasibility of solid fermentation of P. heterophylla fibrous roots was preliminarily evaluated. For in vitro probiotic effect, the increased concentration of P. heterophylla fibrous roots extract dependently increased the total biomass of B. subtilis BD-K010;1.0% P. heterophylla fibrous roots extract received the best effect, and the final p H of 1.0% experimental group was closer to neutral. Meantime, B. subtilis BD-K010 with optimum concentration of P. heterophylla fibrous roots showed significantly higher in vitro antibacterial effect than the control group(P<0.01),and the antibacterial effects on Escherichia coli and Staphylococcus aureus were improved by 51.99% and 63.16%, but it was ineffective to Salmonella. For solid fermentation, the profile of substrate complex and appendage flocculent structure on substrate surface at the end of fermenta-tion in experimental groups added with B. subtilis BD-K010 and cellulase plus BD-K010 were more complex;the live bacteria number, polysaccharide content and saponin content at fermentation end-point in two experimental groups were extremely higher than those in the control group(P<0.01).P. heterophylla fibrous roots extract had good in vitro probiotic effect on B. subtilis BD-K010 and promoted its antibacterial effect, and it is feasible to use probiotics for solid fermentation of P. heterophylla fibrous roots and to improve effective components. 本文研究了不同环境因子和碳氮源对分离自海水养殖环境的一株短小芽孢杆菌(Bacillus pumilus,编号BP-171)无机氮降解特性的影响。环境因子设置了C∶N(质量比)、盐度和pH三种,碳源设为乙酸钠、丁二酸钠、柠檬酸钠、蔗糖、乳糖和葡萄糖,... 本文研究了不同环境因子和碳氮源对分离自海水养殖环境的一株短小芽孢杆菌(Bacillus pumilus,编号BP-171)无机氮降解特性的影响。环境因子设置了C∶N(质量比)、盐度和pH三种,碳源设为乙酸钠、丁二酸钠、柠檬酸钠、蔗糖、乳糖和葡萄糖,氮源设置为氨态氮、亚硝酸态氮、硝酸态氮及三者混合氮源(1∶1∶1)。研究表明,在温度28℃下,菌株BP-171在碳源为葡萄糖、C/N=18、盐度20~40和pH=7.0~8.0条件下对降解液中无机氮有较好的去除作用。在上述脱氮条件下,进一步研究3种单一氮源和1种混合氮源对菌株BP-171无机氮降解特性的影响。研究表明,当氮源为单一的氨态氮、亚硝酸态氮、硝酸态氮时,5天后培养液中氨态氮、亚硝酸态氮、硝酸态氮的去除率最高可分别达74.49%、53.61%和83.39%,总氮则分别降低了2.91%、23.89%和30.98%;在以三者混合作为氮源时,降解液中氨态氮、亚硝酸态氮、硝酸态氮的最高去除率分别为97.89%、78.73%和46.57%,总氮则降低了15.14%。不同条件下培养液中不同形态氮含量的变化情况表明,菌株BP-171对无机氮的转化和去除包括菌体细胞的同化作用、异养硝化作用和好氧反硝化作用等生物学过程,不同条件下具体机制有所差异。 展开更多

Backgrounds In this work, we study two seemingly unrelated aspects of core genetic nonlinear dynamical control of the competence phenotype in Bacillus subtilis, a common Gram-positive bacterium living in the soil. Methods: We focus on hitherto unchartered aspects of the dynamics by exploring the effect of time-scale separation between transcription and translation and, as well, the effect of intrinsic molecular stochasticity. We consider these aspects of regulatory control as two possible evolutionary handles. Results: Hence, using theory and computations, we study how the onset of oscillations breaks the excitability-based competence phenotype in two topologically close evolutionary-competing circuits: the canonical "wild-type" regulation circuit selected by Evolution and the corresponding indirect-feedback inverted circuit that failed to be selected by Evolution, as was shown elsewhere, due to dynamical reasons. Conclusions^ Relying on in-silico perturbation of the living state, we show that the canonical core genetic regulation of excitability-based competence is more robust against switching to phenotype-breaking oscillations than the inverted feedback organism. We show how this is due to time-scale separation and stochasticity. 展开更多

大小兴安岭可培养细菌的资源多样性

细菌作为微生物中的重要组成部分,在有机质的形成与分解、维持生态系统平衡、促进动植物发育等多方面都有着重要作用。2014–2017年,我们采用常见培养基分离纯培养法及16S rRNA基因序列分析方法对大小兴安岭地区土壤可培养细菌的群落结... 细菌作为微生物中的重要组成部分,在有机质的形成与分解、维持生态系统平衡、促进动植物发育等多方面都有着重要作用。2014–2017年,我们采用常见培养基分离纯培养法及16S rRNA基因序列分析方法对大小兴安岭地区土壤可培养细菌的群落结构和多样性进行了调查研究。结果表明:从大小兴安岭地区的17个自然保护区内不同生境的土壤中分离获得3,180个菌株,隶属于24属120种。其中,芽孢杆菌属(Bacillus)的种数和株数最多,分别为38种和2,419株,是大小兴安岭地区可培养细菌的绝对优势类群(占总株数的76.1%);其次是短杆菌属(Brevibacterium)(13.0%)。大兴安岭地区的物种数、Simpson多样性指数和Shannon-Wiener指数高于小兴安岭地区。优势类群芽孢杆菌属的枯草芽孢杆菌(B.subtilis)、苏云金芽孢杆菌(B.thuringiensis)、巨大芽孢杆菌(B.megaterium)等有重要的生产及科学研究价值。 展开更多

Effects of Bacillus subtilis on Degradation of Cellulose 预览

Bacillus subtilis is a representative probiotic widely used in food, medicine, livestock and other industries. In this experiment a strain of Bacillus subtilis was isolated and identified, and its ability to degrade cellulose was also measured. The results showed that Bacillus subtilis had strong capacity to degrade cellulose(62.3% was degraded) and weak capacity to degrade hemicellulose(17.2% was degraded), while it could hardly degrade lignin. The total protein amount in the fermentation medium with cellulose-rich substrate reached by 9.4% after Bacillus subtilis fermentation, compared with that without cellulose-rich substrate. Furthermore, the amounts of Met, Lys and Leu reached by 31.4%, 42.2% and 4.9%, respectively. At 36 h of fermentation, the activity of cellulase reached the highest, and at this time the activity of the enzyme was obtained at 7.19 U · mL-1. The mRNA expression level of the cellulase gene was detected by qPCR, and the experimental group with cellulose substrate was about 2.5 times more than that of the non-cellulose substrate control group. These above results demonstrated that this strain of Bacillus subtilis had a strong ability to degrade cellulose, and synthesize more proteins and restrictive essential amino acids. This study revealed that Bacillus subtilis was a new alternative to ferment cellulose substrates to produce commercial feed or feed additives. 展开更多

Bacillus amyloliquefaciens YP6,a plant growth promoting rhizobacteria,is capable of efficiently degrading a wide range of organophosphorus pesticides(OPs).Here,we report the complete genome sequence of this bacterium with a genome size of 4009619 bp,4210 protein-coding genes and an average GC content of 45.9%.Based on the genome sequence,several genes previously described as being involved in solubilizing-phosphorus,OPs-degradation,indole-3-acetic acid(IAA)and siderophores synthesis.Interestingly,compared with the genomes of B.amyloliquefaciens species,strain YP6 had larger genome size and the most protein-coding genes.Moreover,the four categories of“cell envelope biogenesis,outer membrane(M),”“translation,ribosomal structure and biogenesis(J),”“transcription(K),”and“signal transduction mechanisms(T)”were fewer.These differences may be related to extensive environmental adaptability of the genus B.amyloliquefaciens.These results expand the application potential of strain YP6 for environmental bioremediation,provide gene resources involved in OPs degradation for biotechnology and gene engineering,and contribute to provide insights into the relationship between microorganism and living environment. 展开更多

The demand of silver is increasing rapidly in recent decades, because silver and its related products are widely used in modern industry and decoration. It is necessary to recover silver from waste water using an efficient and environmental friendly method due to its environmental and economic benefits. In this paper, we eliminated the interference of Cl-and light conditions, and then studied the characterization and biosorption performance of silver by Bacillus licheniformis. The max biosorption amount was 87.4 mg/g(dry weight) with the initial Ag+concentration of 100 mg/L at pH 6.0. XRD pattern showed that the product was an amorphous compound. SEM/EDS-mapping and FT-IR results implied that phosphate, amino and carboxyl groups located on the cell walls involved in the biosorption of Ag^+. The XPS spectra result showed that the value of EB of Ag 3d(5/2367.51 eV corresponded to the energy values for Ag(Ⅰ), and indicated Ag^+ adsorbed to the surface of cell still maintained mono-valence. The results confirm that B. licheniformis just adsorb Ag^+ but cannot covert soluble Ag^+ to silver nano-particles(AgNP). 展开更多

为探讨芽孢杆菌和肉桂醛对半滑舌鳎肠道结构的影响,选取60尾初始平均体重为(112±5)g的健康半滑舌鳎,随机分为饲喂普通饲料组(对照组P1)、饲料中含肉桂醛组(P2)、饲料上喷洒益生菌组(P3)、肉桂醛+益生菌组(P4),每组3个重复,每个重复... 为探讨芽孢杆菌和肉桂醛对半滑舌鳎肠道结构的影响,选取60尾初始平均体重为(112±5)g的健康半滑舌鳎,随机分为饲喂普通饲料组(对照组P1)、饲料中含肉桂醛组(P2)、饲料上喷洒益生菌组(P3)、肉桂醛+益生菌组(P4),每组3个重复,每个重复5尾鱼。以工厂化微水流模式养殖,养殖60天后解剖取半滑舌鳎的前肠,制作石蜡切片并以HE染色,观察肠道结构。研究结果表明:饲喂芽孢杆菌使得前肠绒毛出现末端分支、局部增厚,且绒毛中央乳糜管宽度增加。无论肉桂醛、芽孢杆菌还是两者复合使用,均显著降低了前肠的肌层厚度,部分淋巴细胞聚集在黏膜下层呈现炎性浸润,或者迁移到上皮细胞之间。单独使用芽孢杆菌和肉桂醛联用时降低了前肠杯状细胞密度。 展开更多

试验旨在研究混合菌株发酵肉鸡全价配合饲料的条件优化及其饲喂效果。采用四因素三水平的正交试验设计对枯草芽孢杆菌菌株CB-11和植物乳酸杆菌菌株FJ-38在接种量、温度、料水比、发酵时间4个因素对全价料pH值、小肽和干物质回收率(DMR)... 试验旨在研究混合菌株发酵肉鸡全价配合饲料的条件优化及其饲喂效果。采用四因素三水平的正交试验设计对枯草芽孢杆菌菌株CB-11和植物乳酸杆菌菌株FJ-38在接种量、温度、料水比、发酵时间4个因素对全价料pH值、小肽和干物质回收率(DMR)3个指标加权评分影响,对固体发酵全价料进行优化。为进一步验证发酵饲料的饲养效果,进行为期42 d的肉鸡饲养试验。结果表明:①混合菌发酵全价料的最佳工艺条件为:接种量9%、发酵温度39℃、料水比1:1、发酵时间72 h。②10%和15%发酵料添加量显著提高了肉鸡的平均日增重、养分消化率和盲肠中乳酸杆菌和双歧杆菌的数量(P<0.05),降低了料肉比和盲肠中大肠杆菌和沙门氏菌数量(P<0.05)。由此可见,发酵肉鸡全价料在条件优化后营养价值在一定程度得到提高,饲粮添加10%和15%的发酵全价料能提高肉鸡生长性能和养分利用率,改善肠道微生态。 展开更多
