Insecticide toxicity associated with detoxification enzymes and genes related to transcription of cuticular melanization among color morphs of Asian citrus psyllid

The Asian citrus psyllid (Diaphorina citri Kuwayama) is known to exhibit abdominal color polymorphisms. In the current study, susceptibility to four insecticides was compared among orange/yellow, blue/green and gray/brown color morphs of field collected D. citri. The LD50 values and 95% fiducial limits were quantified for each insecticide and color morph combination and ranged between 0.10 ng/μL (0.06-0.10) and 6.16 ng//μL (3.30-12.50). Second, we measured the detoxification enzyme activity levels of orange/yellow, blue/green and gray/brown color morphs for cytochrome P450, glutathione "S-transferase, and general esterase. The mean P450 activity (equivalent units) was significantly lower in gray/brown (0.152 ± 0.006) and blue/green morphs (0.149 ± 0.005) than in the orange/yellow morphs (0.179 ± 0.008). GST activity (/imol/min/mg protein) was significantly lower in the orange/yellow morph (299.70 ± 1.24) than gray/brown (350.86 ± 1.19) and blue/green (412.25 ±1.37) morphs. The mean EST activity (/xmol/min/mg protein) was significantly higher in blue/green (416.72±5.12) and gray/brown morphs (362.19 ± 4.69) than in the orange/yellow morphs (282.56 ± 2.93). Additionally, we analyzed the relative expression of assortment genes involved in cuticular melanization and basal immunity. The transcripts of Dopa Decarboxylase and Tyrosine Hydroxylase were expressed higher in blue/green and gray/brown than orange/yellow morphs. The transcription results paralleled the susceptibility of D.citri to organophosphate, neonicotinoid and pyrethroid insecticides. GST and EST activity may also be correlated with low levels of insecticide susceptibility. Cuticular melanization could be a factor for the development of resistance to insecticides among different color morphs. 展开更多

We investigated the potential hepatoprotective effect of Radix Bupleuri(RB) by inducing acute liver injury(ALI) in an animal model using acetaminophen(APAP) after pretreatment with RB aqueous extract for three consecutive days. Compared to those of the APAP group, the biochemical and histological results of the RB pretreatment group showed lower serum aspartate transaminase(AST) and alanine transaminase(ALT) levels as well as less liver damage. Pharmacokinetic study of the toxicity related marker acetaminophen-cysteine(APC) revealed a lower exposure level in rats, suggesting that RB alleviated APAP-induced liver damage by preventing glutathione(GSH) depletion. The results of cocktail approach showed significant inhibition of CYP2 E1 and CYP3 A activity. Further investigation revealed the increasing of CYP2 E1 and CYP3 A protein was significantly inhibited in pretreatment group,while no obvious effect on gene expression was found. Therefore, this study clearly demonstrates that RB exhibited significant protective action against APAP-induced acute live injury via pretreatment, and which is partly through inhibiting the increase of activity and translation of cytochrome P450 enzymes, rather than gene transcription. 展开更多

Engineering the biosynthesis of plant-derived natural products in microbes presents several challenges, especially when the expression and activation of the plant cytochrome P450 enzyme is required. By recruiting two enzymes—HpaB and HpaC—from several bacteria, we constructed functional 4- hydroxyphenylacetate 3-hydroxylase (4HPA3H) in Saccharomyces cerevisiae to take on a role similar to that of the plant-derived cytochrome P450 enzyme and produce caffeic acid. Along with a common tyrosine ammonia lyase (TAL), the different combinations of HpaB and HpaC presented varied capabilities in producing the target product, caffeic acid, from the substrate, L-tyrosine. The highest production of caffeic acid was obtained with the enzyme combination of HpaB from Pseudomonas aeruginosa and HpaC from Salmonella enterica, which yielded up to (289.4 ± 4.6) mg-L1 in shake-flask cultivation. The compatibility of heterologous enzymes within a yeast chassis was effectively improved, as the caffeic acid production was increased by 40 times from the initial yield. Six key amino acid residues around the flavin adenine dinucleotide (FAD) binding domain in HpaB from Pseudomonas aeruginosa were differentiate from those other HpaBs, and might play critical roles in affecting enzyme activity. We have thus established an effective approach to construct a highly efficient yeast system to synthesize non-native hydroxylated phenylpropanoids. 展开更多

Cytochromes P450 s(CYPs) are terminal enzymes in CYP dependent monooxygenases, which constitute a superfamily of enzymes catalysing the metabolism of both endogenous and exogenous substances. One of their main tasks is to facilitate the excretion of these substances and eliminate their toxicities in most phase 1 reactions. Endogenous substrates of CYPs include steroids, bile acids, eicosanoids, cholesterol, vitamin D and neurotransmitters. About 80% of currently used drugs and environmental chemicals comprise exogenous substrates for CYPs. Genetic polymorphisms of CYPs may affect the enzyme functions and have been reported to be associated with various diseases and adverse drug reactions among different populations. In this review, we discuss the role of some critical CYP isoforms(CYP1 A1, CYP2 D6, CYP2 J2, CYP2 R1,CYP3 A5, CYP3 A7, CYP4 F3, CYP24 A1, CYP26 B1 and CYP27 B1) in the pathogenesis or aetiology of ulcerative colitis concerning gene polymorphisms. In addition, their significance in metabolism concerning ulcerative colitis in patients is also discussed showing a clear underestimation in genetic studies performed so far. 展开更多

The performance of matrix-assisted laser desorption/ionization time of flight mass spectrometry(MALDI-TOF-MS) is frequently compromised by the high limited of detection, heterogeneous distribution of matrix and analyte deposits and time-consuming drying time during the conventional drop drying procedure. Here, we describe a method based on membrane separation technology to assist in the drying of nanomaterials to rapidly make homogeneous sample spots and to enhance the mass signal. We demonstrate that the sample can be dried evenly in a few seconds and MS signal can be greatly optimized by using membrane separation technology. Enrichment with nanomaterials greatly enhances analyte signal strength while membrane separation method avoids the time-consuming requirements of searching for “sweet spots”. 展开更多

This study sought to explore the relationship between cytochrome P450 2C19(CYP2C19)*2/*3 polymorphisms and the development of coronary heart disease(CHD),and to evaluate the influence of the single nucleotide polymorphisms(SNPs)on the occurrence of adverse clinical events in CHD patients.A total of 231 consecutive patients candidate for percutaneous coronary intervention genotyped for CYP2C19*2(681G>A)and*3(636G>A)polymorphisms were enrolled.The adverse clinical events were recorded during a follow-up period of 14 months.The incidence of CHD,according to coronary angiography,was significantly higher(P=0.025)in CYP2C19*2 carriers group.Stepwise binary logistic regression analysis revealed that among factors that potentially influenced the presence of CHD(age>60 years,gender,BMI,etc.),CYP2C19*2 carriers(OR 1.94,95%CI:1.08-3.50,P=0.028)and male gender(OR 2.74,95%CI:1.58-4.76,P=0.001)were independent predictors,which were associated with the presence of CHD.The follow-up results showed that the incidence of adverse cardiovascular events within 14 months of discharge was significantly higher in the CYP2C 19*2 carriers than in the non-carriers(21.6%vs.6.3%,P=0.019).The results of the multivariate Cox proportional hazards model showed that CYP2C19*2 loss-of-flinction was the only independent factor which predicted the coronary events during the follow-up period of 14 months(OR=3.65,95%CI:1.09-12.25,P=0.036).The adverse impact of CYP2C19*2 polymorphisms was found not only in the risk of the presence of CHD,but also in the adverse cardiovascular events in CHD patients during the follow-up period of 14 months.However the same influence was not found in CYP2C19*3 mutation in Chinese Han population. 展开更多

Cytochrome P450 and UDP-glucosyltransferase (UGT) as phase I and phase II metabolism enzymes, respectively, play vital roles in the breakdown of endobiotics and xenobiotics. Insects can in crease the expression of detoxificatio n enzymes to cope with the stress from xenobiotics including insecticides. However, the molecular mechanisms for insecticide detoxification in Spodoptera exigua remain elusive, and the genes conferring insecticide metabolisms in this species are less well reported. In this study, 68 P450 and 32 UGT genes were identified. Phylogenetic analysis showed gene expansions in CYP3 and CYP4 clans of P450 genes and UGT33 family of this pest. P450 and UGT genes exhibited specific tissue expression patterns. Insecticide treatments in fat body cells of S. exigua revealed that the expression levels of P450 and UGT genes were significantly influenced by challenges of abamectin, lambda-cyhalothrin, chlorantraniliprole, metaflumizone and indoxacarb. Multiple genes for detoxification were affected in expression levels after insecticide exposures. The results demonstrated that lambda-cyhalothrin, chlorantraniliprole, metaflumizone and indoxacarb induced similar responses in the expression of P450 and UGT genes in fat body cells;eight P450 genes and four UGT genes were co-up-regulated significantly, and no or only a few CYP/UGT genes were down-regulated significantly by these four insecticides. However, abamectin triggered a distinct response for P450 and UGT gene expression;more P450 and UGT genes were down-regulated by abamectin than by the other four compounds. In con elusion, P450 and UGT genes from S. exigua were identified, and different responses to abamectin suggest a different mechanism for insecticide detoxification. 展开更多

目的观察脓毒症大鼠细胞色素水平和细胞色素C氧化酶活性对肝细胞线粒体生物能量合成和肝脏功能的影响。方法将SD大鼠24只,分为正常对照组(C组)、脓毒症组(S组),每组12只。采用盲肠结扎穿孔(CLP)法建立大鼠脓毒症模型。制模18h... 目的观察脓毒症大鼠细胞色素水平和细胞色素C氧化酶活性对肝细胞线粒体生物能量合成和肝脏功能的影响。方法将SD大鼠24只,分为正常对照组(C组)、脓毒症组(S组),每组12只。采用盲肠结扎穿孔(CLP)法建立大鼠脓毒症模型。制模18h后处死大鼠,取血检测丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)和天冬氨酸氨基转移酶线粒体同工酶(m-AST),使用光镜和电子显微镜分别观察肝细胞和线粒体形态,采用还原-氧化差光谱法测定线粒体细胞色素C、C1、B和AA3含量,测定细胞色素C氧化酶活性,检测线粒体膜电位水平,测定肝细胞三磷酸腺苷(ATP)含量。结果与C组比较,S组血清ALT[(233.02±110.16)U/L]、AST[(742.56±441.41)U/L]和m-AST[(412.78±252.56)U/L]明显升高[C组相应值为(29.81±13.16)、(99.78±41.67)、(46.23±24.24)U/L,P=0.000、0.001、0.001],肝细胞ATP含量[(19.27±8.76)nmol/mg]明显减少[C组(94.65±17.79)nmol/mg,P=0.000],线粒体中细胞色素C[(0.104±0.04)nmol/mg]和AA3[(0.21±0.04)nmo]/mg]明显下降[C组相应值为(0.17±0.03)、(0.38±0.09)nmoL/mg,P=0.000],细胞色素c氧化酶活性[(734.14±107.37)amol/(min·mgprot)]明显下降[c组(1279.75±186.77)nmol/(min·mg prot),P=0.000],线粒体膜电位水平(1.75±0.96)明显下降(C组:3.81±1.16,P=0.000)。结论脓毒症时肝细胞线粒体细胞色素含量减少和细胞色素c氧化酶活性下降,可引起线粒体功能障碍和急性肝细胞搠伤。 展开更多

Objective: To explore the effect of gene polymorphism locus rs12794714 on allergic rhinitis in hainan province, providing reference for genetic diagnosis and prevention. Methods:From March 2017 to December 2017, 94 local patients with Allergic rhinitis in hainan and 113 healthy people who were admitted to the first affiliated hospital of hainan medical college during the physical examination in the physical examination department of hainan medical college were selected. Statistical software SPSS 25.0 was used to conduct inter-group differentiation analysis on genotypes and alleles, and P<0.05 was considered to be statistically different. Results: all the polymorphisms of rs12794714 in hainan population could be detected with the distribution frequency of aa-ag and GG in the Allergic rhinitis group of 20.2%, 46.8%and 33%, and in the control group of 18.6%, 38% and 43.4%. Codominant model, dominant model, recessive model and superdominant model validation were not statistically significant. The distribution frequencies of alleles A and G were 54% and 46% in the experimental group, 63% and 37% in the control group, with no statistical difference. Conclusion: rs12794714 single nucleotide polymorphism has no significant effect on the pathogenesis of Allergic rhinitis in hainan province. 展开更多

Background:Food is an important environmental factor that affects animals’energy metabolism and food shortage has significant effects on animals’behavior,physiology and biochemistry.However,to date few studies have focused on the thermogenesis and its effects on the body condition of birds.In this study,we examined the effects of food restriction on the body mass,basal metabolic rate(BMR)and body composition,and several physiological,biochemical and molecular markers potentially related to thermogenesis,in the Chinese Bulbul(Pycnonotus sinensis).Methods:Birds in the control group were provided with food ad libitum whereas those in the food restriction group were provided with one-half of the usual quantity of food for 12 days.Oxygen consumption was measured using an open-circuit respirometry system.Mitochondrial state 4 respiration and cytochrome c oxidase(COX)activity in the liver and pectoral muscle were measured with a Clark electrode.Avian uncoupling protein(avUCP)mRNA expression was determined in pectorals muscle with quantitative Real-time PCR.Results:Chinese Bulbuls in food restriction group decreased in body mass,BMR and internal organ(heart,kidneys,small intestine and total digestive tract)mass compared with the control group over the 12-day period of food restriction.Bulbuls in the food restriction group also had lower levels of state-4 respiration,COX activity in the liver and muscle,and mitochondrial avUCP gene expression in muscle compared to the control group.BMR was positively correlated with body mass,state 4 respiration in the liver and COX activity in the muscle.Conclusions:Our data indicate that Chinese Bulbuls not only sustain food shortage through simple passive mechanisms,such as reducing body and organ mass and energy expenditure,but also by reducing energetic metabolism in the liver and muscle. 展开更多

AIM To evaluate the impact of cytochrome P450 2C19(CYP2C19)and interleukin-1β(IL-1β)polymorphisms on the efficacy of Helicobacter pylori(H.pylori)eradication by using rabeprazole-based hybrid therapy.METHODS A total of 88 H.pylori-infected patients were recruited to receive 14-d of hybrid therapy from March 2013 to May 2014.Three patients were excluded from analysis because of incomplete compliance.Either a follow-up endoscopy or 13C-urea test was performed to determine the results of H.pylori eradication therapy.The genotypes of CYP2C19 and IL-1βwere analyzed to investigate the impact on treatment effect.RESULTS The total eradication rate of H.pylori was 92.94%(79/85).According to the CYP2C19 genotypes,the rates of H.pylori eradication were 89.19%in extensive metabolizers(EM)and 95.83%in non-EM.The H.pylori eradication rates regarding the IL-1βgenotypes were 92.59%in the normal acid secretion group and 93.10%in the low acid secretion group.After multivariable logistic regression analysis,both the genotypes of CYP2C19 and IL-1βhad no significant influences on the eradication rates of H.pylori.CONCLUSION The CYP2C19 and IL-1βpolymorphisms are not significantly independent factors of H.pylori eradication using rabeprazole-based hybrid therapy. 展开更多

In China, it was first reported from Hainan Province, and later from several other provinces. In the present study, a rice root-knot nematode population found from the rice cultivation areas of Zhejiang Province, China is characterized via molecular analysis using internal transcribed spacer(ITS) and cytochrome c oxidase subunit Ⅱ(coxⅡ)-16 S rRNA genes and scanning electron microscopy(SEM) observations of males and the second-stage juveniles. Morphometric data and molecular sequence comparisons for all M. graminicola populations occurring in China are also provided. The overall morphology of M. graminicola found in Zhejiang match well with the original description, though males have a slightly longer body and stylet, and a shorter tail, while the second-stage juvenile is also slightly longer than in the original description. This is the first report of M. graminicola from Zhejiang. Phylogenetic studies based on coxⅡ suggest that all the Chinese populations belong to Type B. This study expands knowledge of the increasing distribution and phylogenetic relationships of M. graminicola that occur in China. 展开更多

The expression level of cytochrome P450 genes in insects can be induced by plant allelochemicals,which is important for insects to adapt to host plants.Cytochrome P450CYP6B 7has been reported to be involved in pyethroid insecticide resistance in Heli- coverpa armigera,and its transcription level was induced by some inducers.Currently,the regulatory mechanism of the induced expression of CYP6B7remains unknown,although it is very important for understanding the detoxification mechanism to allelochemicals in host plants.The objective of the present study was to investigate the eis-acting ele- ment in the promoter of CYP6B7 mediating the inducible up-regulation of CYP6B7in H.armigera by 2-tridecanone.The promoter region of CYP6B7was cloned by genome walking technique and analyzed by transient transfeetion assay.Progressive 5'deletion of the promoter region of CYP6B7revealed that the relative luciferase activity of construct -320/+232could be significantly induced by 2-trideeanone.Further stepwise deletion between -320 and -238 bp found that construct -292/+232 could also be significantly induced by 2-tridecanone,but the adjacent construct -256/+232could not,suggesting the essential role of the sequence between -292 and --257 bp for 2-tridecanone induction. Nucleotide mutations between -292 and -281 bp had no influence on the induction ef- fect by 2-tridecanone,but nucleotide mutations between -280 and -257 bp significantly decreased the induction effect.These results demonstrated that the cis-acting element for 2-trideeanone induction was between -280 and -257 bp in the promoter of CYP6B7. 展开更多

Cyp19a1b、Cyp11a1、Cyp11b2、Cyp17a1、Hsd3b1等细胞色素相关基因能够调节硬骨鱼类性类固醇的合成,对性腺发育和性别决定产生影响。本研究以全雌三倍体虹鳟(Oncorhynchus mykiss)为研究对象,正常雌性二倍体虹鳟为对照,选取31~68 dpf(days post fertilization)时间段的虹鳟仔鱼脑组织,采用qRT-PCR和酶联免疫的方法研究以上几种基因的表达状况和脑芳香化酶的活性变化,以期探明导致三倍体雌性虹鳟性腺发育异常的关键原因。qRTPCR结果显示,二倍体中Cyp19a1b在30~50 dpf时表达量上调并且维持在较稳定水平,但50~56 dpf时表达量逐渐下调,之后56~68 dpf表达量持续上调;三倍体中Cyp19a1b表达量在30~35 dpf开始上调,35~47 dpf逐渐下调,47~55 dpf开始第二次上调,之后维持在较稳定水平直至68 dpf,但三倍体Cyp19a1b的表达量显著(P<0.05)低于同期二倍体的。二倍体Cyp11a1表达量在34 dpf出现峰值,三倍体Cyp11a1在38 dpf时出现峰值。二倍体Hsd3b1表达量在33~42 dpf时维持在较高水平,在38 dpf时出现高峰;三倍体Hsd3b1表达量在47~59 dpf时较高,在49 dpf出现高峰。二倍体中Cyp11b2在37 dpf出现峰值,之后开始下调;三倍体在40 dpf出现峰值,之后逐渐下调,但三倍体Cyp11b2表达量显著低于同期二倍体。二倍体Cyp17a1的表达量在35~46 dpf时逐渐上升,在45 dpf时达到高峰之后直至69 dpf逐渐下降,并且维持在较为平稳的水平上;但是在相同的实验条件下未检测到同一时期三倍体Cyp17a1的表达量。酶联免疫结果显示,在40 dpf时二者的脑芳香化酶活性到达高峰,但在40~60 dpf时期,二倍体虹鳟脑芳香化酶活性显著(P<0.05)高于三倍体虹鳟,尤其在45~50 dpf时,该酶活性分别较三倍体的高1.15倍和1.12倍。以上结果表明三倍体虹鳟早期性腺发育迟缓的原因之一是Cyp19a1b、Cyp11a1、Cyp11b2、Cyp17a1、Hsd3b1等基因的表达晚于二倍体,且表达量低于二倍体,造成雌二醇不能正常合� 展开更多

The cytochrome bc1 complex(the bc1 complex or complex Ⅲ) is an attractive target for the discovery of numerous pharmaceuticals and pesticides. In order to identify new lead structures for this target, a new series of molecules, N-(4-aryloxyphenyl)phthalimides, were designed and synthesized in a straightforward manner. Our design strategy was to introduce a 4-aryloxyphenyl group, a fragment which exhibited promising bc1 complex-inhibiting properties, into the aryl group of the valuable N-arylphthalimide backbone. Afterward, the biochemical evaluation of the newly synthesized compounds was carried out,and the results implied that several compounds demonstrated good activities against succinatecytochrome reductase(SCR, a mixture of mitochondrial complex Ⅱ and the bc1 complex). Further studies confirmed that 3e’, a representative compound in this paper, was identified as an inhibitor of the bc1 complex. Furthermore, computational simulations were also performed to better understand binding of 3e’ to the enzyme complex, which indicated that 3e’ should bind to the Qo site of the bc1 complex.Consequently, we harbor the idea that this paper can provide a solid platform for synthesis and discovery of other bc1 complex inhibitors. 展开更多
