目的探讨中药炮制对含苷类药物疗效和不良反应的影响。方法选取辽宁省盘锦市中心医院2017年5月至2019年5月收治的160例接受含苷类药物治疗的患者作为研究对象,随机分为两组,对照组80例患者接受未炮制的含苷类药物治疗,试验组80例患者接... 目的探讨中药炮制对含苷类药物疗效和不良反应的影响。方法选取辽宁省盘锦市中心医院2017年5月至2019年5月收治的160例接受含苷类药物治疗的患者作为研究对象,随机分为两组,对照组80例患者接受未炮制的含苷类药物治疗,试验组80例患者接受经过炮制的含苷类药物治疗,比较两组临床疗效、不良反应发生率、生命质量评分。结果临床治疗有效率试验组(97.50%)明显高于对照组(81.25%),试验组不良反应发生率(2.50%)明显低于对照组(16.25%),试验组生命质量评分明显高于对照组,差异有统计学意义(P<0.05)。结论中药炮制可显著提高含苷类药物的临床治疗效率,降低不良反应发生率,提高患者生命质量。 展开更多

Lianqiao(Forsythia suspensa(Thunb.)Vahl,Abbreviated as“F.suspensa”below)is traditional Chinese medicine in China,widely distributed all over the country and also highly rich in resources.The Northwest Territories is a genuine producing area.There are many chemical components in F.suspensa,including lignans,phenylethanolic glycosides,flavonoids,C6-C2 natural alcohols and their glycosides,phenolic acids,terpenes and volatile oils.Among them,lignans and phenylethanolic glycosides are the main active components.Based on the review of chemical components or constituents and main pharmacological activities,according to the definition of quality marker,the components of quality marker of F.suspensa were predicted and analyzed from the aspects of plant genetics and chemical composition,traditional efficacy,traditional medicine,new efficacy,blood entering composition,measurable composition,effective composition in different compatibility,and the influence of storage conditions of extract.Further research on its medicinal active ingredients will provide scientific basis for the evaluation of F.suspensa quality. 展开更多

苗药铁筷子不同炮制品中4种成分含量及毒性的比较研究 认领

目的:比较苗药铁筷子阴干、烘干、蒸制、炒炭制品中4种成分的含量及其毒性。方法:采用高效液相色谱法(HPLC)同时测定铁筷子炮制品中东莨菪苷、秦皮苷、东莨菪内酯、异嗪皮啶的含量。色谱柱为Welch-C18,流动相为乙腈-水(梯度洗脱),流速为... 目的:比较苗药铁筷子阴干、烘干、蒸制、炒炭制品中4种成分的含量及其毒性。方法:采用高效液相色谱法(HPLC)同时测定铁筷子炮制品中东莨菪苷、秦皮苷、东莨菪内酯、异嗪皮啶的含量。色谱柱为Welch-C18,流动相为乙腈-水(梯度洗脱),流速为1 mL/min,检测波长为214 nm,柱温为30℃,进样量为10μL。采用改良寇氏法测定铁筷子不同加工炮制品对小鼠的半数致死量(LD50)及其95%置信区间(CI),并评价其急性毒性。结果:东莨菪苷、秦皮苷、东莨菪内酯、异嗪皮啶检测质量的线性范围分别为0.019~1.856、0.016~1.616、0.009~0.920、0.006~0.624μg(R2均不低于0.9995),精密度、稳定性、重复性试验的RSD均小于2.0%(n=6);平均加样回收率分别为104.49%、102.22%、101.45%、99.26%(RSD均小于2%,n=6);阴干、烘干、蒸制、炒炭品中,东莨菪苷含量分别为1.1190%、0.9043%、1.0684%、0.0364%,秦皮苷含量分别为0.8678%、0.4539%、0.4237%、0.0205%,东莨菪内酯含量分别为0.0771%、0.1555%、0.0844%、0.2908%,异嗪皮啶含量分别为0.1102%、0.2021%、0.2081%、0.2494%;4种炮制品的LD50分别为4118.13、3733.36、1643.61、>10000 mg/kg,95%CI分别为(3748.87,4523.76)、(3422.16,4072.86)、(1520.90,1776.23)、(>10000)mg/kg。结论:HPLC法重复性、精密度较好,可用于同时测定铁筷子不同炮制品中4种成分的含量;不同炮制品中4种成分均有所差异,2个糖苷类毒性成分的含量明显下降;4种炮制品均为低毒或无毒。 展开更多

OBJECTIVE:To investigate the antioxidant property and potential ameliorating effective ingredients for high altitude-induced fatigue from Gansu Maxianhao(Pedicularis Kansuensis Maxim.).METHODS:Macroporous adsorptive resin combined with polyamide chromatographic column was used to obtain water extract(P1),high polar part(P2),iridoid glycosides part(P3)and phenylethanoid glycosides part(P4)of Gansu Maxianhao(Pedicularis Kansuensis Maxim.).Antioxidant activity of each part was investigated employing a series of in vitro models.Hematoxylin-eosin staining,analysis of blood biochemical parameters,along with molecular analyses examining oxidative stress makers,metabolite,metabolic enzyme and energy substance in liver,skeletal muscle and/or serum were further measured.RESULTS:The results showed phenylethanoid glycosides(Ph Gs)exhibited more effective with 1,1-diphenyl-2-picrylhydrazyl radical scavenging ability,2-2-Azinbis(3-ehtylbenzothiazolin-6-sulfnic Acid)Diammonium Salt+scavenging activity,hydroxyl radical scavenging activity,reducing ability and antioxidant activity to varying extents.Under a hypobaric hypoxia attack in a hypobaric chamber(8000 m,7 consecutive days),Ph Gs was administered to BALB/C mice at doses of 50,200,400 mg/kg and antifatigue property was evaluated using a swimming test at an altitude of 4000 m.The results showed that Ph Gs of Gansu Maxianhao(Pedicularis Kansuensis Maxim.)could significantly prolong the burden swimming time of mice,reduce the hypoxia-induced oxidative stress,remove the accumulated products of metabolism,improve the energy metabolism as well as improve preservation of endogenous glycogen stores.CONCLUSION:The ameliorating effect against altitude-induced fatigue of Ph Gs from Gansu Maxianhao(Pedicularis Kansuensis Maxim.)might come from the alleviation of oxidative stress,reduction of the adverse metabolic products,normalizing energy metabolism and increasing energy substances reserves.Ph Gs is a potential antioxidant and novel remedy for fatigue due to high-altitude hypoxia. 展开更多

Brewers who create hop-forward styles such as American-style India Pale Ales typically add hops toward the end of or after wort boiling to avoid aroma volatilization and thereby impart strong hopderived aromas.However,previous studies have demonstrated that despite the volatilization effects of boiling wort,hops that are added early in the kettle boil can contribute to hop aroma.Nonvolatile hopderived precursors,specifically glycosides,may survive the boiling process and become hydrolyzed to release volatile aglycones capable of contributing to aroma.To investigate the contribution of these different hop fractions to the aroma of kettle-hopped beers,12 single-hopped pilot-scale(3 hL)beers were brewed using pellet,supercritical CO2 extract,and spent hop fractions of Citra,Simcoe,Centennial,or Cascade cultivars.Pellet,extract,and spent additions consisted of a single hop add让ion 5 min into a 60 min boil.Volatile analysis of beers was perfonned using stir-bar sorptive extraction and quantified using gas chromatographymass spectrometry.Beers were analyzed for the common terpenoid compoundsα-pinene,β-pinene,3-myrcene,limonene,linalool,E,β-caryophyllene,α-humulene,andα-terpineol.In addition,beers were evaluated using descriptive sensory analysis.The descriptive sensory data identified significant differences among the cultivar and hop product treatments.The spent hop treatments produced beers that had a noticeable hop aroma,which suggests that the water-soluble components left behind in the spent hops may contribute to hop aroma in beer.The intensity and nature of the hop aroma in the spent treatments were hop variety specific.However,the contributions of water-soluble components found in spent hops to increased aroma intensity in beer were small,especially compared with the pellet and extract treatments. 展开更多

Introduction::Tripterygium glycosides (TGs) have been widely used in China to treat diabetic nephropathy (DN);however, proof of their use is scarce. The present study aimed to evaluate the effectiveness and safety of adding TGs to angiotensin-converting enzyme (ACE) inhibitors or angiotensin receptor blockers (ARBs).Methods::By searching Embase, MEDLINE, Cochrane Library, SINOMED, China National Knowledge Infrastructure, VIP Information/Chinese Scientific Journals, and WANFANG databases, we identified previous studies that met the specific selection criteria and included them in the meta-analysis. Analyses were performed using Review Manager (version 5.3).Results::Nine randomized controlled trials were included in the final meta-analysis. Patients were compared before and after treatment with ACE inhibitors or ARBs plus TGs, or ACE inhibitors or ARBs alone. The results revealed that treatment with ACE inhibitors or ARBs plus TGs resulted in significantly greater reductions in 24-h urinary total protein (UTP) levels (trial duration <2 months, mean difference [MD]: -0.25;95% confidence interval [ CI]: -0.32, -0.18;trial duration between 2 and 6 months, MD: -0.39;95% CI: -0.44, -0.33;trial duration >6 months, MD: -2.09;95% CI: -2.89, -1.29) compared with treatment using ACE inhibitors or ARBs alone. Additionally, ACE inhibitors or ARBs plus TGs showed better results after longterm administration. Treatment with ACE inhibitors or ARBs plus TGs resulted in significantly greater reductions in serum creatinine (SCr) compared with ACE inhibitors or ARBs alone (MD: -9.87;95% CI: -13.76, -5.97). Conclusion::In patients with DN, adding TGs to ACE inhibitors or ARBs significantly lowered both the 24-h UTP and SCr levels. Therefore, ACE inhibitors or ARBs plus TGs might improve the treatment of DN in patients. 展开更多

目的建立薄层色谱法、高效液相色谱(HPLC)法对玄参中多种苷类成分进行同时检测,为简便快速评价玄参药材质量提供参考依据。方法建立薄层色谱法对玄参药材的质量进行初步评价;采用高效液相色谱-二极管阵列检测器(HPLC-PDA)法检测苷类成... 目的建立薄层色谱法、高效液相色谱(HPLC)法对玄参中多种苷类成分进行同时检测,为简便快速评价玄参药材质量提供参考依据。方法建立薄层色谱法对玄参药材的质量进行初步评价;采用高效液相色谱-二极管阵列检测器(HPLC-PDA)法检测苷类成分含量,WatersSymmetry-C18(250 mm×4.6 mm,5μm)色谱柱;柱温:25℃;以乙腈(A)-水(B)为流动相进行梯度洗脱;流速1.0 mL·min-1;检测波长分别为:哈巴苷、桃叶珊瑚苷和麦角甾苷为206 nm,哈巴俄苷为280 nm,安格洛苷C为320 nm。结果供试品的薄层色谱中,在与对照药材和对照品色谱相应的位置上,显相同颜色的斑点。HPLC法测定的20批次玄参药材中,除麦角甾苷外其他主要苷类成分基本能稳定存在,但含量差异较大;通过对几种苷类成分的总量进行对比分析发现,道地药材浙玄参具有较明显优势,湖南新邵基地的玄参药材中苷类成分总量具有较高的一致性,提示作为玄参生产基地,湖南新邵生产的玄参药材质量较稳定。结论所建立的薄层色谱法和HPLC法能快速对玄参中多种苷类成分进行鉴定和含量测定,可为玄参药材等级标准的制定提供参考。 展开更多

Objective:To investigate the therapeutic effect of total iridoid glycosides of Picrorhiza scrophulariiflora(TIGP)on non-alcoholic steatohepatitis(NASH).Methods:SD rats were fed with high-fat and high-sugar diet for 8 weeks to establish NASH.TIGP were given orally at doses of 20,40 and 80 mg/kg/d for 4 weeks.Triglycerides assay(TG),total cholesterol(TC),low density lipoprotein cholesterol(LDL-C),high-density lipoprotein cholesterol(HDL-C),aspartate aminotransferase(AST),alanine aminotransferase(ALT),fasting plasma glucose(FPG),fasting insulin(FINS),tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),chemokine-1(MCP-1),leptin(LEP)in serum were tested.TG,TC,superoxide dismutase(SOD),malondialdehyde(MDA),and free fatty acid(FFA)in liver tissue were determined by colorimetric methods.Steatosis of hepatocytes and inflammation was performed by pathological examination.Results:The results showed that TIGP significantly decreased TC,TG and FFA in liver tissue,increased SOD activity,decreased MDA content,decreased serum levels of TG,TC,HDL-C/LDL-C,ALT,AST,GLU,HOMA-IR,TNF-αand LEP,and in addition,improved steatosis of liver cells compared to NASH.Conclusion:TIGP had anti-fatty liver effect against NASH rats induced by high-fat and high-sugar diet.Its mechanism was related to the regulation of lipid metabolism and reduction of insulin resistance,through inhibition of oxidative stress and inflammation. 展开更多

The present work sought to characterize the cyanogenic glycoside dhurrin in five sugarcane varieties and to determine its possible relationship with the formation of ethyl carbamate in cachaca. For each variety, methanol, ethyl acetate and hexane extracts were prepared and submitted to thin layer chromatography. Chromatographic spots were revealed with iodine vapors. The physical and chemical parameters of the spirits produced from the five different varieties of sugar cane and by different production processes were within the legal limits. Although not exceeding the limit for ethyl carbamate, a significant variation in the final concentration of this compound was observed for each sample analyzed. The Rf values for the dhurrin standard and for the sugarcane variety SP 83-2847 were equal, similar to those for SP 80-3280 and CTC 11 and different from the Rf values for the RB 86-7515 and IAC 86-2480 samples. 展开更多

Combination therapy of tripterygium glycosides plus valsartan in diabetic nephropathy treatment: A systematic review and meta-analysis 认领

Objective: To assess the efficacy and safety of the combination therapy of Chinese herbal medicines,tripterygium glycosides, plus valsartan for the treatment of diabetic nephropathy(DN).Methods: A comprehensive research of 12 electronic databases was performed. Data of the included studies were extracted and analyzed independently by two authors, and were synthesized using Review Manager(version 5.2) and Stata(version 12.0).Results: A total of 12 randomized controlled trials(RCTs) involving 829 patients were included. Pooled results showed that the combination therapy significantly increased total efficacy of DN patients [RR = 1.35,95% CI(1.22, 1.50), P < 0.001], the level of serum albumin [Mean difference(MD)= 3.87, 95% CI(3.12,4.62), P < 0.001], and significantly decreased 24 h urinary protein level [MD =-0.97, 95% CI(-1.19,-0.76), P < 0.001], urinary albumin-excretion rate [MD =-145.53, 95% CI(-227.95,-63.11), P < 0.001],and urinary β2-microglobuli level [MD =-11.86, 95% CI(-13.02,-10.69), P < 0.001]. No significant differences were found in levels of serum creatinine [MD =-0.26, 95% CI(-7.52, 7.00), P > 0.05), blood urea nitrogen [MD = 0.25, 95% CI(-0.23, 0.74), P > 0.05], and endogenous creatinine clearance rate[MD =-0.43, 95% CI(-3.48, 2.62), P > 0.05]. However, tripterygium glycosides plus valsartan seemed to exert higher adverse reaction rate than valsartan monotherapy [MD = 3.41, 95% CI(1.34, 8.66), P < 0.05].There were no publication bias for all of the pooled effect sizes.Conclusion: Combination therapy of tripterygium glycosides plus valsartan may be effective for the treatment of DN. However, the safety of the combination therapy need to be further confirmed. 展开更多

Objective:Dichocarpum auriculatum,an endangered perennial herb,is endemic to China and has been used as folk medicines for the treatment of cough,hepatitis,scrofula,and epilepsy.However,there is no phytochemical report on this herbal so far.For the resource development and protective importance of this endangered medicinal plant,a phytochemical study was undertaken.Methods:The chemical constituents were purified by silica gel column,Sephadex LH-20 column,and semi-preparative reversed phase HPLC.NMR and MS spectra were used for structural identification.Results:Thirteen compounds were isolated from D.auriculatum.Their structures were characterized as jatrorrhizine(1),berberine(2),steponine(3),magnoflorine(4),coclauril(5),menisdaurin(6),menisdaurilide(7),aquilegiolide(8),(6 R,9 S)-3-oxo-α-ionol-β-D-glucopyranoside(9),blumenol C glucoside(10),palmitic acid(11),dibutylphthalate(12),and auriculatum A(13).Conclusion:Compound 13 is a new diester terephthalate derivative.All the compounds are obtained from the genus Dichocarpum for the first time,and compounds 9 and 10 have potential chemotaxonomic significance to the genus Dichocarpum. 展开更多

Objective: To summarize the evidence from systematic reviews(SRs) on the benefits and safety of Tripterygium glycosides(TG) and total glucosides of paeony(TGP), commonly used to treat rheumatoid arthritis(RA) in China, for patients with RA. Methods: SRs of randomized controlled trials(RCTs) on TG or TGP in treating RA were included, by searching 8 databases from their inception until December 2017. Two authors extracted data independently. We assessed the quality of SRs using AMSTAR and graded the quality of evidence according to the GRADE approach. Results: Eleven SRs containing an average of 7.6 RCTs, involving a total of 7,012 participants were included in this overview. On the basis of included SRs, TG and TGP could improve the following indexes for RA patients: American College of Rheumatology(ACR) 20 response rate, ACR50 response rate and ACR70 response rate, swollen joint count, tender joint count, erythrocyte sedimentation rate and C-reactive protein. Moreover, TGP could reduce incidence of hepatotoxicity. The most common adverse effects of TG were gastrointestinal discomfort and gonad toxicity, while for TGP was mild to moderate diarrhea. The overall quality of evidence for these findings ranged from "low" to "moderate". Conclusions: TG and TGP might be 2 potentially effective complementary and alternative drugs for patients with RA. Nevertheless, due to gonad toxicity, TG should only be considered in elderly patients or patients without reproductive needs. More evidence from high quality RCTs and SRs is warranted to support the use of TG and TGP for RA patients. 展开更多

Flaxseed cake contains cyanogenic glucosides, which can be metabolized into hydrocyanic acid in an animal’s body, leading to asphyxia poisoning in cells. Beta-glucosidase is highly efficient in degrading cyanogenic glucosides. The Cattle may have b-glucosidase-producing strains in the intestinal tract after eating small amounts of flaxseed cake for a long time. This study aimed to isolate of a strain from cow dung that produces b-glucosidase with high activity and can significantly reduce the amount of cyanogenic glucosides. We used cow dung as the microflora source and an esculin agar as the selective medium. After screening with 0.05% esculin and 0.01% ferric citrate, we isolated 5 strains producing high amounts of b-glucosidase. In vitro flaxseed cake fermentation was fermented by these 5 strains, in which the strain M-2 exerted the best effect(P < 0.05). The strain M-2 was identified as Lichtheimia ramosa and used as the fermentation strain to optimize the fermentation parameters by a single factor analysis and orthogonal experimental design. The optimum condition was as follows: inoculum size3%, water content 60%, time 144 h, and temperature 32℃. Under this condition, the removal rate of cyanogenic glucosides reached 89%, and crude protein increment reached 44%. These results provided a theoretical basis for the removal of cyanogenic glucosides in flaxseed and the comprehensive utilization of flaxseed cake. 展开更多

As Garcinia Glycosides is an effective but insoluble anti-tumor drug, the aim of this study was to obtain a solid dispersion form Garcinia Glycosides by using solvent-melt method so that improve the solubility and dissolution rate. The solid dispersion was characterized by High Performance Liquid Chromatography (HPLC), infrared spectroscopy and evaluated the intestinal absorption of the drug by rat in situ single pass intestinal perfusion. The results showed the increase of solubility, dissolution velocity and absorption compared to other forms. This indicated that solid dispersion could greatly improve the relative bioavailability of Garcinia Glycosides in vivo. 展开更多
