Zebrafish and human genomes are highly homologous;however,despite this genomic similarity,adult zebrafish can achieve neuronal proliferation,regeneration and functional restoration within 6–8 weeks after spinal cord injury,whereas humans cannot.To analyze differentially expressed zebrafish genes between axon-regenerated neurons and axon-non-regenerated neurons after spinal cord injury,and to explore the key genes and pathways of axonal regeneration after spinal cord injury,microarray GSE56842 was analyzed using the online tool,GEO2R,in the Gene Expression Omnibus database.Gene ontology and protein-protein interaction networks were used to analyze the identified differentially expressed genes.Finally,we screened for genes and pathways that may play a role in spinal cord injury repair in zebrafish and mammals.A total of 636 differentially expressed genes were obtained,including 255 up-regulated and 381 down-regulated differentially expressed genes in axon-regenerated neurons.Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment results were also obtained.A protein-protein interaction network contained 480 node genes and 1976 node connections.We also obtained the 10 hub genes with the highest correlation and the two modules with the highest score.The results showed that spectrin may promote axonal regeneration after spinal cord injury in zebrafish.Transforming growth factor beta signaling may inhibit repair after spinal cord injury in zebrafish.Focal adhesion or tight junctions may play an important role in the migration and proliferation of some cells,such as Schwann cells or neural progenitor cells,after spinal cord injury in zebrafish.Bioinformatic analysis identified key candidate genes and pathways in axonal regeneration after spinal cord injury in zebrafish,providing targets for treatment of spinal cord injury in mammals. 展开更多

Beta-nerve growth factor(β-NGF)is known to be a major leading cause of neuronal plasticity.To identify the possible action mechanisms ofβ-NGF gene therapy for sciatic nerve recovery,experimental dogs were randomly divided into control,pyridoxine,and pyridoxine+β-NGF groups.We observed chronological changes of morphology in the dorsal root ganglia in response to pyridoxine toxicity based on cresyl violet staining.The number of large neurons positive for cresyl violet was dramatically decreased after pyridoxine intoxication for 7 days in the dorsal root ganglia and the neuron number was gradually increased after pyridoxine withdrawal.In addition,we also investigated the effects ofβ-NGF gene therapy on neuronal plasticity in pyridoxine-induced neuropathic dogs.To accomplish this,tyrosine kinase receptor A(TrkA),βIII-tubulin and doublecortin(DCX)immunohistochemical staining was performed at 3 days after the last pyridoxine treatment.TrkA-immunoreactive neurons were dramatically decreased in the pyridoxine group compared to the control group,but strong TrkA immunoreactivity was observed in the small-sized dorsal root ganglia in this group.TrkA immunoreactivity in the dorsal root ganglia was similar betweenβ-NGF and control groups.The numbers ofβⅢ-tubulin-and DCX-immunoreactive cells decreased significantly in the pyridoxine group compared to the control group.However,the reduction ofβⅢ-tubulin-and DCX-immunoreactive cells in the dorsal root ganglia in theβ-NGF group was significantly ameliorated than that in the pyridoxine group.These results indicate thatβ-NGF gene therapy is a powerful treatment of pyridoxine-induced neuropathic damage by increasing the TrkA and DCX levels in the dorsal root ganglia.The experimental protocol was approved by the Institutional Animal Care and Use Committee(IACUC)of Seoul National University,South Korea(approval No.SNU-060623-1,SNU-091009-1)on June 23,2006 and October 9,2009,respectively. 展开更多

Objective To determine whether relative abundance of epidermal growth factor receptor (EGFR) mutations in plasma predicts clinical response to epidermal growth factor receptor-tyrosine kinase inhibitors (EGFR-TKIs) in patients with advanced lung adenocarcinoma. Methods In this prospective study,adult patients with advanced lung adenocarcinoma were enrolled in our hospital from 1 April 2016 to 1 January 2017. EGFR mutations in tumortissues were detected by ADx-amplification refractory mutationsystem (ADx-ARMS). EGFR mutations of plasmafree tumor DNA were detected by ADx-ARMS and ADxsuperamplification refractory mutation system (ADx-SuperARMS)at the same time. Patients with EGFR-mutantin tumor tissues and receiving EGFR-TKIs were finallyenrolled. Plasma mutation-positive patients with bothmethods were high abundance group. Patients with positivemutations by ADx-SuperARMS but negative of ADx-ARMS were medium abundance group. 展开更多

Pirfenidone suppresses the abnormal activation of human Müller cells after platelet-derived growth factor-BB stimulation

AIM: To determine the effect of pirfenidone on the activated human Müller cells by platelet-derived growth factor-BB(PDGF-BB). METHODS: The primary human Müller cells were separated from retinal tissues and established the pathogenic model by stimulated with PDGF-BB. The Müller cells behaviour of normal group and the model group was measured by MTT assay, Trypan blue assay, cell migration assay, and collagen contraction assay. The expression of transforming growth factor(TGF)-β1,-β2, and pigment epithelium-derived factor(PEDF) was estimated with realtime polymerase chain reaction(PCR), Western blot and immunofluorescence analyses. RESULTS: A pathogenic/proliferative model of Müller cells was established by stimulating normal cultured Müller cells with 10 ng/mL PDGF-BB for 48 h. After treated with 0.2 and 0.3 mg/mL pirfenidone, the proliferation, migration and collagen contraction was statistically significantly depressed in the model group compared with the normal groups. The expression levels of TGF-β1 and TGF-β2 were significantly down-regulated, while the PEDF expression was significantly up-regulated after treated with 0.2 and 0.3 mg/mL pirfenidone in the model group. CONCLUSION: Pirfenidone effectively suppress the proliferation, migration and collagen contraction of the human Müller cells stimulated with PDGF-BB through down-regulation of TGF-β1/TGF-β2 and up-regulation of PEDF. 展开更多

The role of vascular endothelial growth factor A in platelet adhesion in cerebral microvessels in the early stage of subarachnoid hemorrhage remains unclear.In this study,the endovascular puncture method was used to produce a rat model of subarachnoid hemorrhage.Then,30 minutes later,vascular endothelial growth factor A antagonist anti-vascular endothelial growth factor receptor 2 antibody,10μg,was injected into the right ventricle.Immunohistochemistry and western blot assay were used to assess expression of vascular endothelial growth factor A,occludin and claudin-5.Immunohistochemical double labeling was conducted to examine co-expression of GP Ⅰa-Ⅱ integrin and type Ⅳ collagen.TUNEL was used to detect apoptosis in the hippocampus.Neurological score was used to assess behavioral performance.After subarachnoid hemorrhage,the expression of vascular endothelial growth factor A increased in the hippocampus,while occludin and claudin-5 expression levels decreased.Co-expression of GP Ⅰa-Ⅱ integrin and type Ⅳ collagen and the number of apoptotic cells increased,whereas behavioral performance was markedly impaired.After treatment with anti-vascular endothelial growth factor receptor 2 antibody,occludin and claudin-5 expression recovered,while co-expression of GP Ⅰa-Ⅱ integrin and type Ⅳ collagen and the number of apoptotic cells decreased.Furthermore,behavioral performance improved notably.Our findings suggest that increased vascular endothelial growth factor A levels promote platelet adhesion and contribute to early brain injury after subarachnoid hemorrhage.This study was approved by the Biomedical Ethics Committee,Medical College of Xi’an Jiaotong University,China in December 2015. 展开更多

Safety and efficacy of EFGR and VEGF signaling pathway inhibition therapy in patients with colorectal cancer: a meta-analysis 预览

However, few studies have focused on the safe... Objective Epidermal growth factor receptor(EGFR) and vascular endothelial growth factor(VEGF) inhibitors are two targeted therapies for metastatic colorectal cancer(mCRC). However, few studies have focused on the safety and efficacy of combined targeted therapy against those of a single inhibition therapy of EFGR or VEGF. This meta-analysis aimed to compare the anti-tumor activity of the combined inhibition therapy and single inhibition therapy in patients with mCRC. Methods We searched PubMed, Medline, the Cochrane library, Embase, and annual meeting proceedings for relevant clinical trials. Objective response rate(ORR), progression-free survival(PFS), overall survival(OS), and adverse events were extracted and calculated.Results Nine trials comprising 3977 patients were selected for the analysis. The combined inhibition therapy showed a 3.7% improvement in ORR compared with single inhibition, and this difference was statistically significant [hazard ratio(HR)= 1.33;95% confidence interval(CI), 1.01–1.74;P = 0.04]. Subgroup analysis showed that the combined EGFR and VEGF inhibitor therapy had an 11.65% improvement in ORR compared with VEGF inhibitor therapy(OR = 2.14;95% CI, 1.34–3.40;P = 0.001). EGFR and VEGF inhibitor therapy and chemotherapy had an 18.08% improvement in ORR compared with chemotherapy(OR = 2.21;95% CI, 1.05–4.64;P = 0.04). Moreover, EGFR and VEGF inhibitor therapy significantly improved PFS compared with VEGF inhibitor therapy(OR = 0.82;95% CI, 0.69–0.97;P = 0.02). VEGF inhibitor therapy and chemotherapy significantly improved PFS compared with EGFR and VEGF inhibitor therapy and chemotherapy(OR = 1.20;95% CI, 1.11–1.30;P = 0.00). In addition, EGFR and VEGF inhibitor therapy showed improved OS compared with VEGF inhibitor therapy(HR = 0.78, 95% CI: 0.65–0.94;P = 0.008). OBJECTIVE:To determine the therapeutic effect and potential mechanism of Huatan Tongluo decoction on rats with collagen-induced arthritis.METHODS:Forty specific pathogen-free Wistar rats were selected,and 10 were randomly selected as the control(group 1).The remaining rats were injected intradermally with emulsified type II bovine collagen at the tail base and back,followed by a booster 7 d post first immunization.After establishing collagen-induced arthritis(CIA),rats were randomly divided into three groups(n=10).The rats were treated orally for 30 d as follows:group 1,saline;group 2,model(saline);group 3,tripterygium polyglycoside(TP;7.81 mg/kg,positive control);group 4,Huatan Tongluo decoction(HTTL;7.5 g/kg).Body weight,ankle swelling and arthritis index were measured over the course of the study.The rats were sacrificed 30 d after treatment.Morphological changes in the synovium were observed by hematoxylin and eosin staining.Pannus formation and synovial thickness in the left ankle were observed by color Doppler ultrasoundVascular endothelial growth factor(VEGF)and VEGFR2 protein levels were measured by immunohistochemistry.VEGF/VEGFR2 mRNA levels were measured by real-time quantitative polymerase chain reaction.RESULTS:Compared with the model group,a significantly lower arthritis index was observed in the positive control group(P<0.05)and HTTL group(P<0.01),after treatment.Both positive control and HTTL reduced intra-articular pannus formation and synovial thickening.Furthermore,VEGF mRNA,and VEGFR2 protein and mRNA levels were significantly downregulated(P<0.05)in the treatment groups.CONCLUSION:Inhibition of the expression of VEGF and VEGFR2 in synovial tissues and the formation of pannus and synovial hyperplasia may be part of the mechanism of HTTL for relieving the symptoms of rheumatoid arthritis in CIA rats. 展开更多

Hepatocellular carcinoma(HCC)is the fifth most common cancer,and hepatitis C virus(HCV)infection plays a major role in HCC development.The molecular mechanisms by which HCV infection leads to HCC are varied.HCV core protein is an important risk factor in HCV-associated liver pathogenesis and can modulate several signaling pathways involved in cell cycle regulation,cell growth promotion,cell proliferation,apoptosis,oxidative stress and lipid metabolism.The dysregulation of signaling pathways such as transforming growth factorβ(TGF-β),vascular endothelial growth factor(VEGF),Wnt/β-catenin(WNT),cyclooxygenase-2(COX-2)and peroxisome proliferator-activated receptorα(PPARα)by HCV core protein is implicated in the development of HCC.Therefore,it has been suggested that this protein be considered a favorable target for further studies in the development of HCC.In addition,considering the axial role of these signaling pathways in HCC,they are considered druggable targets for cancer therapy.Therefore,using strategies to limit the dysregulation effects of core protein on these signaling pathways seems necessary to prevent HCV-related HCC. 展开更多

目的观察人脐带间充质干细胞对糖尿病足溃疡模型大鼠血清炎性因子及相关细胞生长因子的影响,探讨其治疗糖尿病足溃疡的作用机制.方法 50只无特定病原体(SPF)级雄性大鼠使用高脂饮料喂养4周后,采用腹腔注射1%链脲佐菌素(40 mg/kg)、水蒸... 目的观察人脐带间充质干细胞对糖尿病足溃疡模型大鼠血清炎性因子及相关细胞生长因子的影响,探讨其治疗糖尿病足溃疡的作用机制.方法 50只无特定病原体(SPF)级雄性大鼠使用高脂饮料喂养4周后,采用腹腔注射1%链脲佐菌素(40 mg/kg)、水蒸汽熏蒸的方法建立糖尿病足溃疡模型,取建模成功30只大鼠分为模型组、高剂量人脐带间充质干细胞治疗组(高剂量治疗组:股动脉注射人脐带间充质干细胞2.0×10^6/0.2 ml)、低剂量人脐带间充质干细胞治疗组(低剂量治疗组:股动脉注射人脐带间充质干细胞1.0×10^6/0.2 ml)各10只,另取10只SPF级雄性大鼠大鼠为空白对照组(空白组).比较溃疡面积愈合率、血清炎性因子、相关细胞生长因子、肉芽组织成纤维细胞数及新生毛细血管数等指标.应用SPSS 21.0统计软件分析,符合正态分布的计量资料用均值±标准差(Mean±SD)表示,多组间比较采用方差分析,两两比较采用配对t检验,不符合正态分布的计量资料用四分位数间距表达,多组间比较采用Kruskal-WallisH检验,两组比较采用Manne Whitnery U检验.结果(1)创面愈合率:模型组、低剂量治疗组、高剂量治疗组分别为(26.31±4.32)%、(66.92±7.24)%、(74.74±8.32)%.低剂量治疗组、高剂量治疗组创面愈合率明显高于模型组(t=8.424、11.201,P<0.01),高剂量治疗组创面愈合率明显高于低剂量治疗组(t=3.148,P<0.05).(2)血清炎性因子:模型组、低剂量治疗组、高剂量治疗组血清白细胞介素-1(IL-1)、肿瘤坏死因子-α(TNF-α)、超敏C-反应蛋白(hs-CRP)含量分别为(6.12 ±0.25)、(4.54±0.36)、(4.20±0.33) ng/L;(29.45±3.12)、(21.13±3.24)、(18.20±2.65) ng/L;(0.61±0.08)、(0.45±0.07)、(0.37±0.06) ng/L.低剂量治疗组、高剂量治疗组大鼠血清IL-1、TNF-α、hs-CRP含量明显低于模型组(t=11.400、5.849、5.111、14.666、8.691、8.222,P<0.05,P<0.01);高剂量治疗组大鼠IL-1、TNF-α、hs-CRP含 展开更多

Furthermore, current clinical treatments do not yet approach the best effectiveness. Thus, the development of better strategies for treating diabetic neuropathy is an urgent matter. In this review, we first discuss the advantages and disadvantages of some major mouse models of diabetic neuropathy and then address the targets for mechanism-based treatment that have been studied. We also introduce our studies on each part. Using stem cells as a source of neurotrophic factors to target extrinsic factors of diabetic neuropathy, we found that they present a promising treatment. 展开更多

AIM: To evaluate the effect of β-elemene on the expressions of hypoxia-inducible factor(HIF)-lα, vascular endothelial growth factor(VEGF) and inducible nitric oxide synthase(i NOS) in a streptozotocin(STZ) induced diabetic SpragueDawley(SD) rat model.METHODS: SD rats were administered an abdominal injection of STZ and induced to a diabetic model. After 6 wk course of diabetes, the treatment groups were given β-elemene through periocular and intravitreous injection separately and the control groups were given blank emulsion injection. HE staining was used to observe the morphology of retina. The m RNA expressions of HIF-1α, VEGF and i NOS was assayed by real-time polymerase chain reaction(PCR) and the protein expression was measured by Western blot and immunocytochemistry methods.RESULTS: The results indicated that the protein and m RNA expressions of HIF-1α, VEGF and i NOS after treated by β-elemene periocularly and intravitreally injections were all found to be reduced compared with the levels in the diabetic rats group(P<0.05). The inhibitory effect of intravitreal injection was more remarkable.CONCLUSION: The results show β-elemene protect the retina of diabetic rats from high glucose damage by downregulating the expression of HIF-1α, VEGF and iNOS. 展开更多

Objective:To compare the effects of laparoscopic surgery and traditional open surgery on serum growth factor, immunoglobulin and related factors in patients with hepatocellular carcinoma after operation.Methods: 60 patients with hepatocellular carcinoma admitted to our hospital from 2014 to 2018 were randomly divided into the control group and the observation group, with 30 cases in each group. Patients in the control group underwent traditional open radical hepatectomy, while patients in the observation group underwent laparoscopic radical hepatectomy. Before and after the operation, the levels of Alpha-alanine aminotransferase (ALT), aspartate aminotransferase (AST), vascular endothelial growth factor (VEGF), acidic fibroblast growth factor (aFGF), basic fibroblast growth factor (bFGF), immunoglobulin A (IgA), immunoglobulin M (IgM), immunoglobulin G (IgG), matrix metalloproteinase-13 (MMP-13), Procalcitonin (PCT) and C-reactive protein (CRP) were compared between the patients in the two groups.Results:The levels of ALT, AST, VEGF, aFGF, bFGF, IgA, IgM, and IgG of patients in the control group were significantly decreased after the operation, but the levels of MMP-13, PCT and CRP increased significantly. The serum levels of ALT, AST, VEGF, aFGF, bFGF, IgA, IgM, and IgG of patients in the observation group were significantly decreased after operation, but the levels of MMP-13, PCT and CRP increased significantly. The above indexes of patients in the observation group were better than those in the control group (P<0.05).Conclusion: Laparoscopic hepatectomy has a good clinical effect. It can significantly reduce the level of growth factor in serum of patients. It has less damage to immune function than open surgery and can reduce the level of inflammatory stress caused by operation. 展开更多

Uncoupling neurotrophic function from nociception of nerve growth factor:what can be learned from a rare human disease? 预览

Nerve growth factor(NGF) is a powerful trophic factor that provides essential support for the survival and differentiation of sympathetic and sensory neurons during development. Nerve growth factor(NGF) is a powerful trophic factor that provides essential support for the survival and differentiation of sympathetic and sensory neurons during development. However, NGF also activates nociceptors contributing significantly to inflammatory pain and neuropathic pain after tissue injury. As such anti-NGF based therapies represent a promising strategy for pain management. Because of dose-dependent serious side effects such as back pain, injection site hyperalgesia, clinical trials of using NGF to treat various disorders such as diabetic neuropathies, chemotherapy-induced and human immunodeficiency virus-associated peripheral neuropathies were all discontinued. Thus far, worldwide clinical applications of NGF in treating patients are very limited except in China. Hereditary sensory autonomic neuropathy type V(HSAN V) is an extremely rare disease. Genetic analyses have revealed that HSAN V is associated with autosomal recessive mutations in NGF. One of the mutations occurred at the 100th position of mature NGF resulting in a change of residue from arginine to tryptophan(R100W). Although those HSAN V patients associated with the NGFR100W mutation suffer from severe loss of deep pain, bone fractures and joint destruction, interestingly patients with the NGFR100W mutation do not show apparent cognitive deficits, suggesting important trophic support function is preserved. We believe that NGFR100W provides an ideal tool to uncouple the two important functions of NGF: trophic versus nociceptive. Studies from investigators including ourselves have indeed confirmed in animal testing that the NGFR100W no longer induced pain. More importantly, the trophic function seemed to be largely preserved in NGF harboring the R100W mutation. On the mechanistic level, we found that the NGFR100W mutation was capable of binding to and signaling through the tyrosine receptor kinase A receptor. BACKGROUND Inflammatory cytokines play a vital role in the occurrence of osteoarticular injury and inflammation. Whether inflammation-associated factors interleukin-1β(IL- 1β), IL-6, tumor necrosis factor-α(TNF-α) and vascular endothelial growth factor (VEGF) are involved in the pathogenesis of keen articular cartilage injury remains poorly understood. AIM To measure the levels of inflammatory factors [IL-1β, IL-6, TNF-α and VEGF] in patients with knee articular cartilage injury. METHODS Fifty-five patients with knee articular cartilage injury were selected as patient groups, who were divided into three grades [mild (n = 20), moderate (n = 19) and severe (n = 16)] according to disease severity and X-ray examinations. Meanwhile, 30 healthy individuals who underwent physical examination were selected as the control group. The levels of IL-1β, IL-6, TNF-α and VEGF were measured by ELISA and immunohistochemical staining. RESULTS Compared with the control group, patient groups displayed significantly higher levels of IL-1β, IL-6, TNF-α and VEGF, and the extent of increase was directly proportional to the severity of injury (P < 0.05). In addition, the number of cells with positive staining of IL-1β, IL-6, TNF-α and VEGF in the synovial membrane were significantly increased, along with increased disease severity (P < 0.05). After treatment, the scores of visual analogue scale and the Western Ontario and McMaster University of Orthopaedic Index in patient groups were 2.26 ± 1.13 and 15.56 ± 7.12 points, respectively, which were significantly lower than those before treatment (6.98 ± 1.32 and 49.48 ± 8.96). Correlation analysis suggested that IL-1β and TNF-α were positively correlated with VEGF. CONCLUSION IL-1β, IL-6, TNF-α and VEGF levels are increased in patients with knee articular cartilage injury, and are associated with the disease severity, indicating they might play an important role in the occurrence and development of knee articular cartilage injury.
