Plant distribution and pharmacological activity of flavonoids 预览

Flavonoids are natural organic compounds that are widely found in nature, their structural types are complex, and they mainly include flavonoids, flavonols, dihydroflavonols, isoflavones, dihydroisoflavones, chalcones... Flavonoids are natural organic compounds that are widely found in nature, their structural types are complex, and they mainly include flavonoids, flavonols, dihydroflavonols, isoflavones, dihydroisoflavones, chalcones, orange ketones, flavanoids, anthocyanidins, and biflavonoids. This review covers the plant distribution and pharmacological activities of flavonoids. Flavonoids are mainly distributed in angiosperms and gymnosperms, and they are abundant in plants such as Rutaceae, Labiatae, Zingiberaceae, Scrophulariaceae, and Leguminosae. Because of their wide distribution and variety, researchers have found that flavonoids have diverse biological activities, mainly focusing on anti-inflammatory, antibacterial and antitumor activities. Mechanistically, the anti-inflammatory effects are mainly related to the NF-κB and MAPK (mitogen-activated protein kinase) signaling pathway and then the inhibition of the production of inflammatory cytokines and mediators. The antibacterial activity is mainly manifested as inhibitory effects on many strains, including Escherichia coli, Cryptococcus neoformans, and Pseudomonas aeruginosa, via destroying the stability of the microbial membrane, inhibiting the invasion of virulent bacteria into host cells, promoting the apoptosis of bacteria, inhibiting bacterial fatty acid synthesis, etc. The antitumor activity of flavonoids is related to their inhibition of cell proliferation and induction of apoptosis via the mitochondria-mediated, endoplasmic reticulum-mediated, and death factor and its receptor-mediated signal transduction pathways. Understanding the plant distribution and pharmacological activity of flavonoids not only reveals the importance of identifying such valuable flavonoids in another genus or family but also provides a basis for fully exploiting the therapeutic potential of flavonoids. 展开更多

Stem cell transplantation has brought new hope for the treatment of neurological diseases.The key to stem cell therapy lies in inducing the specific differentiation of stem cells into nerve cells.Because the differentiation of stem cells in vitro and in vivo is affected by multiple factors,the final differentiation outcome is strongly associated with the microenvironment in which the stem cells are located.Accordingly,the optimal microenvironment for inducing stem cell differentiation is a hot topic.EGb761 is extracted from the leaves of the Ginkgo biloba tree.It is used worldwide and is becoming one of the focuses of stem cell research.Studies have shown that EGb761 can antagonize oxygen free radicals,stabilize cell membranes,promote neurogenesis and synaptogenesis,increase the level of brain-derived neurotrophic factors,and replicate the environment required during the differentiation of stem cells into nerve cells.This offers the possibility of using EGb761 to induce the differentiation of stem cells,facilitating stem cell transplantation.To provide a comprehensive reference for the future application of EGb761 in stem cell therapy,we reviewed studies investigating the influence of EGb761 on stem cells.These started with the composition and neuropharmacology of EGb761,and eventually led to the finding that EGb761 and some of its important components play important roles in the differentiation of stem cells and the protection of a beneficial microenvironment for stem cell transplantation. 展开更多

Objective: The 3-N-butylphthalide (NBP) comprises one of the chemical constituents of celery oil. It has a series of pharmacologic mechanisms including reconstructing microcirculation, protecting mitochondrial function, inhibiting oxidative stress, inhibiting neuronal apoptosis, etc. Based on the complex multi-targets of pharmacologic mechanisms of NBP, the clinical application of NBP is increasing and more clinical researches and animal experiments are also focused on NBP. The aim of this review was to comprehensively and systematically summarize the application of NBP on neurologic diseases and briefly summarize its application to non-neurologic diseases. Moreover, recent progress in experimental models of NBP on animals was summarized. Data sources: Literature was collected from PubMed and Wangfang database until November 2018, using the search terms including "3-N-butylphthalide,""microcirculation,""mitochondria,""ischemic stroke,""Alzheimer disease,""vascular dementia,""Parkinson disease,""brain edema,""CO poisoning,""traumatic central nervous system injury,""autoimmune disease,""amyotrophic lateral sclerosis,""seizures,""diabetes,""diabetic cataract," and "atherosclerosis." Study selection: Literature was mainly derived from English articles or articles that could be obtained with English abstracts and partly derived from Chinese articles. Article type was not limited. References were also identified from the bibliographies of identified articles and the authors’ files. Results: NBP has become an important adjunct for ischemic stroke. In vascular dementia, the clinical application of NBP to treat severe cognitive dysfunction syndrome caused by the hypoperfusion of brain tissue during cerebrovascular disease is also increasing. Evidence also suggests that NBP has a therapeutic effect for neurodegenerative diseases. It is being used traditionally to overcome various medical complications like sore eyes, aches, rheumatism, allergy, itching, and rashes. Besides, Acacia auriculiformis has been proven for many pharmacological activities like central nervous system depressant activity, antioxidant, antimicrobial, antimalarial, anti-filarial, cestocidal, antimutagenic, chemopreventive, spermicidal, wound healing, hepatoprotective and antidiabetic activity due to its low toxicity(LD50 = 3 741.7 mg/kg) and high efficacy. In addition, various phytochemical investigations reveal the presence of chief constituents as flavonoids(Auriculoside) and triterpenoid saponin glycosides(acaciasides-acaciaside A & B) in different parts of this plant. Since many years researchers have been carrying out various studies on this medicinal important shrub to elicit the various biological activities. This review attempts to highlight the pharmacognostical, phytochemical and pharmacological observations from 1965 to 2018 retrieved from SciF inder, Scientific journals, books, Google Scholar, and botanical electronic database websites. The various plant extracts evaluated for different pharmacological activities showed significant efficacy. Bioactive phytoconstituents isolated from various parts of the plant are highlighted. Pharmacognostical standardization of the plant done with various standard parameters is also reported. The low toxicity of this plant and the presence of major bioactive phytoconstituents like flavonoids and triterpenoid saponin glycosides are responsible for a therapeutic remedy for various diseases and pharmacological activities respectively. Since many signal transduction and molecular events are implicated in these diseases, a novel method to interrogate the key pharmacological mechanisms is required to accelerate innovative drug discovery. Much attention now focuses on the real-time visualization of molecular disease events to yield new insights to the pathogenesis of the diseases. This review focuses on recent advances in the development of chemical probes for imaging pathological events to facilitate the study of the underlying pharmacodynamics and toxicity involved. As reviewed here, optical imaging is now frequently viewed as an indispensable technique in the field of biological research. Promoting interdisciplinary collaboration among chemistry, biology and medicine, is necessary to further refine functional fluorescent probes for diagnostic and therapeutic applications. 展开更多

A total of 201 compounds from Danshen have been reported, including lipophilic diterpenoids, water-soluble phenolic acids, and other constituents, which have showed various pharmacological activities, such as anti-inflammation, anti-oxidation, anti-tumor, anti-atherogenesis, and anti-diabetes. Ham., a rhizomatous perennial herb of the Saxifragaceae family, has been listed in rare species. In traditional medicine the plant has been used for the treatment of ulcer, bleeding during child birth, inflammation, body ache, diarrhea, and dysentery. This article reviews and analyses the traditional use, pharmacological activities of the corresponding bioactive compounds, and toxicological data on A. rivularis species. A. rivularis has been used across Indian subcontinent to treat various human ailments. Its extract consists of alkaloids, flavonoids, coumarins and glycosides. These extracts as well as isolated bioactive compounds demonstrated various biological activities including antimicrobial, anti-peptic ulcer, anti-inflammatory, and antioxidant activity. Toxicological studies of the extract on brine shrimp larvae demonstrated mild toxicity;however no acute toxicity in mice. Although the pharmacological effect of A. rivularis has been poorly explored, the available data validated its use in ethnomedicine. To reveal the cryptic therapeutic potential of the species, research should focus on identification and mechanistic studies of the bioactive compounds. 展开更多

As a well-known Chinese medicine prescription, Wenxin Keli (WXKL) has been widely used in the treatment of many arrhythmias including atrial fibrillation and has achieved significant clinical effects, but the specific mechanism of its treatment of atrial fibrillation remains unclear. This study used a network pharmacology approach to reveal the potential molecular mechanisms of WXKL in the treatment of atrial fibrillation. It involves oral bioavailability screening of ingredients, drug-likeness evaluation, compound target fishing, H-C-T network and C-T-P network construction and analysis. Through the network pharmacology-based research strategy, the complex system of WXKL was analyzed. It was found that 14 representative compounds of WXKL, 30 protein targets and 12 related signaling pathways help us systematically understand the underlying mechanism of WXKL’s anti-atrial fibrillation effects. 展开更多

Objective: Eryngium with the 274 accepted species, is the largest genus of Apiaceae family which are distributed all over the world and have been used in traditional remedies to manage various ailments in different nations. Ten species of Eryngium have been identified in Iran including E. caeruleum M.B.(syn: E. caucasicum Trautv.), E. creticum Lam., E. bungei Boiss., E. billardieri F. Delaroche.(syn: E. kotschyi Boiss.), E. glomeratum Lam.(syn: E. parviflorum Sm.), E. bornumulleri Nab., E. pyramidale Boiss.& Husson., E. noeanum Boiss., E. wanaturi Woron.(syn: E. woronowii Bordz.), and E. thyrsoideum Boiss. The aim of the present research is to review pharmacological activity, and phytochemical constituents as well as ethnobotany and traditional uses of Iranian species of Eryngium. Materials and methods: Electronic databases including PubMed, Scopus, Science Direct (ISI Web of Knowledge) and Embase library were comprehensively searched for research on Eryngium. The search period was from 1966 to October 2018. The related articles were selected according to the inclusion and exclusion criterias in our study. Results: A total of 57 papers were enrolled in analyses. The findings showed that Iranian species of Eryngium, had a noticeable diverse of traditional medicinal uses and also broad range of pharmacological activities as well as various phytochemical compounds. Some remarkable biological and pharmacological activities of these species have been demonstrated in present scientific studies, including antimicrobial, cytotoxic and anticancer, anti-inflammatory, analgesic and antinociceptive activities as well as antioxidant, antidiabetic, anti-snake and anti-scorpion venom effects. Conclusion: Iranian Eryngium species have enormous potential for prospective preparation of herbal medicinal products and are good candidates for discovering new drugs. 展开更多

The development of new proton-pump inhibitors (PPIs) with less adverse effects by lowering the pKa values of nitrogen atoms in pyrimidine rings has been previously suggested by our group. In this work, we proposed that new PPIs should have the following features:(1) number of ring II = number of ring I+ 1;(2) preferably five, six, or seven-membered heteroatomic ring for stability;and (3) 1<pKa1<4. Six molecular scaffolds based on the aforementioned criteria were constructed, and R groups were extracted from compounds in extensive data sources. A virtual molecule dataset was established, and the pKa values of specific atoms on the molecules in the dataset were calculated to select the molecules with required pKa values. Drug-likeness screening was further conducted to obtain the candidates that significantly reduced the adverse effects of long-term PPI use. This study provided insights and tools for designing targeted molecules in silico that are suitable for practical applications. 展开更多

Plants are known to possess plenty of pharmacological activities as a result of various phytoconstituents. Tetramethylpyrazine(TMP), one of the most widely used medicinal compound isolated from traditional Chinese herb, is usually employed for anti-oxidation, anti-inflammation, anti-platelet aggregation, antilipid, anti-fibrosis, as well as activating blood, removing stasis, dilating small arteries, improving microcirculation and antagonizing calcium. In the present paper, the anti-adhesion effect of TMP were reviewed. TMP was found to play a multi-target and muti-link role in anti-adhesion by inhibiting hyperplasia of collagen and overexpression of adhesion-related factors and reducing the concentration of white blood cells and fibrin in plasma. Because previous studies mostly focused on in vitro experiments and animal experiments, there is an urgent need for clinical research with abundant indicators to further prove its anti-adhesion potency. Future basic research should concentrate on the development of TMP as a biological material. 展开更多
