Gene expression regulation, including loss-of-function and gain-of-function assays, is a powerful method to study developmental and disease mechanisms. Drosophila melanogaster is an ideal model system particularly well-equipped with many genetic tools. In this review, we describe and discuss the gene expression regulation techniques recently developed and their applications, including the CRISPR/Cas9-triggered heritable mutation system, CRISPR/dCas9-based transcriptional activation(CRISPRa) system,and CRISPR/dCas9-based transcriptional repression(CRISPRi) system, as well as the next-generation transgenic RNAi system. The main purpose of this review is to provide the fly research community with an updated summary of newly developed gene expression regulation techniques and help the community to select appropriate methods and optimize the research strategy. 展开更多

目的探讨慢病毒高毒性物质稳定转基因技术对雄性Wistar大鼠生殖系统的影响。方法构建PAX2稳定干扰的含有绿色荧光蛋白的慢病毒,将该病毒溶液用台盼蓝标记,通过34 G针头注入16只Wistar大鼠睾丸输出小管。2周后用超声监测睾丸实质,1个月... 目的探讨慢病毒高毒性物质稳定转基因技术对雄性Wistar大鼠生殖系统的影响。方法构建PAX2稳定干扰的含有绿色荧光蛋白的慢病毒,将该病毒溶液用台盼蓝标记,通过34 G针头注入16只Wistar大鼠睾丸输出小管。2周后用超声监测睾丸实质,1个月后将大鼠处死,取出睾丸及其附属腺体,免疫荧光染色及HE染色观察组织结构形态。结果大鼠睾丸输出小管注射,共有8只注射成功,均出现不同程度的睾丸曲精小管凋亡坏死,部分小管腔内可见报告基因的高表达,附睾上皮未出现明显的转染情况,而精囊腺上皮细胞出现不同程度的绿色荧光蛋白高表达。结论睾丸和附睾不适合雄性动物的慢病毒转染,并且该处转染容易影响精子的成熟和发育,而精囊腺注射慢病毒建立转基因动物模型具有可行性。 展开更多

Porcine viral diarrhea is an acute and highly contagious enteric disease of pigs that causes huge economic losses worldwide. Porcine epidemic diarrhea virus(PEDV) and transmissible gastroenteritis virus(TGEV) are the main pathogens responsible for piglet viral diarrhea. However, currently there is no speci?c drug available for the effective treatment of viral diarrhea.Therefore, it is necessary to seek an effective method to diminish PEDV and TGEV infection rates. RNA interference has been applied successfully to inhibit the virus replication. It provides a potential strategy for breeding resistant pigs. In this study, four promoters and four short hairpin RNA(shRNA) vectors with LoxP sites at each end of the selectable marker genes were constructed to target PEDV and TGEV. These vectors were then transfected into porcine fetal ?broblasts, G418 resistant transfectants were con?rmed by PCR and transgenic SCNT porcine blastocysts were obtained. These results have paved the way for future production of marker-free transgenic resistant to PEDV and TEGV pigs by SCNT. 展开更多

Humankind draws important benefits from large-scale ecological processes termed ecosystem services, yet the status of several of them is declining. Reliable monitoring methods are essential for tracking the status of ecosystem services. Predation is the mainstay of natural pest control, a key ecosystem service. We used green plasticine caterpillars to monitor predation pressure, and to obtain baseline data on predator activity in transgenic Bt versus non-Bt maize fields in Old and New World countries. Predation pressure was measured at ground and canopy levels using an identical, small-plot experimental design in four European countries (Denmark, Slovakia, Romania and Italy) and Argentina. Total predation rate in maize was l l.7%d^-1 (min. 7.2%d^-1 in Argentina, max. 29.0%d^-1 in Romania). Artificial caterpillars were attacked both by invertebrates (mostly chewing insects with 42.0% of the attack marks, and ants with 7.1%, but also predatory and parasitoid wasps, spiders and slugs), and vertebrates (small mammals 25.5%, and birds 20.2%). Total predation at ground level (15.7%d^-1) was significantly higher than in maize canopies (6.0%d^-1) in all countries, except Argentina. We found no significant differences between predator pressure in Bt versus non-Bt maize plots. The artificial caterpillar method provided comparable, quantitative data on predation intensity, and proved to be suitable for monitoring natural pest control. This method usefully expands the existing toolkit by directly measuring ecological function rather than structure. 展开更多

Spermatogonial stem cells(SSCs)transmit genetic information to the next progeny in males.Thus,SSCs are a potential target for germ I i ne modifications to gen erate tran sgenic an imals.In this study,we report a technique for the gen erati on of tran sgenic rats by in vivo manipulation of SSCs with a high success rate.SSCs in juvenile rats were transduced in vivo with high titers of lentivirus harbori ng enhan ced green fluoresce nt protei n and mated with wild-type females to create foun der rats.These founder rats expressed the transgene and passed on the transgene with an overall success rate of 50.0%.Subsequent generations of progeny from the founder rats both expressed and passed on the transgene.Thus,direct modification of SSCs in juvenile rats is an effective means of generating transgenic rats through the male germline.This technology could be adapted to larger animals,in which existing methods for gene modificatio n are in adequate or in applicable,resulti ng in the gen eration of tran sge nic an imals in a variety of species. 展开更多

Homeobox A10(Hoxa10) gene is one of the most important candidate genes associated with the reproductive performance of humans and mice. Overexpression of Hoxa10 in mouse endometrium can increase litter size. Moreover, Hoxa10 plays a key role in regulating the embryo implantation of sows. This study aimed to generate transgenic pigs using Hoxa10 via somatic cell nuclear transfer(SCNT). We established seven Hoxa10-transgenic cell lines, and two of the cell lines were selected as nuclear donors for the transfer. A total of 1 270 cloned embryos were generated and transferred to five surrogate mothers(Landrace×Yorkshire). Eight cloned male piglets were produced including one with cryptorchidism. Six transgenic piglets grew up healthy and produced 56 offspring. Finally, we obtained six transgenic male pigs and 26 transgenic positive offspring that can be used to further study the regulatory mechanism of Hoxa10 on the reproductive performance of pigs. 展开更多

【目的】明确灌木柳碱性α-半乳糖苷酶基因(Sl SIP)的结构特征和该基因在耐盐方面的生物学功能,为碱性α-半乳糖苷酶基因新的功能拓宽提供理论基础。【方法】以灌木柳苏柳‘2345’(Salix jiangsuensis‘2345’)盐胁迫48 h后的c DNA为模... 【目的】明确灌木柳碱性α-半乳糖苷酶基因(Sl SIP)的结构特征和该基因在耐盐方面的生物学功能,为碱性α-半乳糖苷酶基因新的功能拓宽提供理论基础。【方法】以灌木柳苏柳‘2345’(Salix jiangsuensis‘2345’)盐胁迫48 h后的c DNA为模板,克隆了苏柳碱性α-半乳糖苷酶基因Sl SIP。运用生物信息学方法分析其蛋白质结构、性质和亲缘关系,利用农杆菌介导法转入拟南芥中,通过实时荧光定量qRT-PCR方法鉴定转基因阳性植株的表达特性和盐胁迫条件下的生长状态,从而验证转基因植株的耐盐功能。【结果】该基因编码776个氨基酸,分子质量为84.88 ku,理论等电点为5.85,不稳定系数为35.74,亲水性平均系数-0.154,定位于内质网膜上。SlSIP的保守结构域为WFGWCTW和IDDGWQ,催化活性位点为V。共得到11个T3代阳性转基因株系,qRT-PCR结果显示Sl SIP在3个拟南芥阳性转基因株系中表达量最高,分别提高9、28、29倍。转基因株系在含85 mmol/L Na Cl培养基上种子萌发率高于非转基因植株,且生长状态良好,但非转基因植株的叶绿素含量高于转基因株系。【结论】Sl SIP基因属于GH27家族,不仅提高了种子萌发过程中的耐盐性,也参与了叶片发育和衰老的途径。 展开更多

The widespread planting of insect-resistant crops has caused a dramatic shift in agricultural landscapes, thus raising concerns about the potential impacts on both target and non-target pests. In this study, we examined the potential effects of intra-specific seed mixture sowing with transgenic Bt rice (Bt) and its parental non-transgenic line (Nt)(100% Bt rice [Bt100], 5% Nt+95% Bt [Nt05Bt95], 10% Nt+90% Bt [Nt|0Bt90], 20% Nt+80% Bt [Nt2oBtso], 40% Nt+60% Bt [Nt4oBt6O] and 100% Nt rice [Ntjoo]) on target and non-target pests in a 2-year field trial in southern China. The occurrence of target pests, Sesamia inf evens. Chilo suppressalis and Cnaphalocrocis medinalis, decreased with the increased ratio of Bt rice, and the mixture ratios with more than 90% Bt rice (Btjoo and Nt()5Bt95) significantly increased the pest suppression efficiency, with the lowest occurrences of non-target planthoppers, Nilaparvata lugens and Sogatella jurcijera in Ntioo and NtosBtgs. Furthermore, there were no significant differences in 1000?grain dry weight and grain dry weight per 100 plants between Btjoo and NtosBtgs,Seed mixture sowing of Bt rice with <10%(especially 5%) of its parent line was sufficient to overcome potential compliance issues that exist with the use of block or structured refuge to provide most effective control of both target and non-target pests without compromising the grain yield. It is also expected that the strategy of seed mixture sowing with transgenic Bt rice and the nontransgenic parental line would provide rice yield stability while decreasing the insecticide use frequency in rice production. 展开更多

Wheat high-molecular-weight glutenin subunits(HMW-GS)determine dough elasticity and play an essential role in processing quality.HMW-GS are encoded by Glu-1 genes and controlled primarily at transcriptional level,implemented through the interactions between cis-acting elements and trans-acting factors.However,transcriptional mechanismof Glu-1 genes remains elusive.Here we made a comprehensive analysis of cis-regulatory elements within 1-kb upstream of the Glu-1 start codon(-1000 to-1)and identified 30 conserved motifs.Based on motif distribution pattern,three conserved cis-regulatory modules(CCRMs),CCRM1(-300 to-101),CCRM2(-650 to-400),and CCRM3(-950 to-750),were defined,and their functions were characterized in wheat stable transgenic lines transformed with progressive 5′deletion promoter::GUS fusion constructs.GUS staining,qPCR and enzyme activity assays indicated that CCRM2 and CCRM3 could enhance the expression level of Glu-1,whereas the 300-bp promoter(-300 to-1),spanning CCRM1 and core region(-100 to-1),was enough to ensure accurate Glu-1 initiation at 7 days after flowering(DAF)and shape its spatiotemporal expression pattern during seed development.Further transgenic assays demonstrated that CCRM1-2(-300 to-209)containing CompleteHMWEnhancer(-246 to-209)was important for expression level but had no effect on expression specificity in the endosperm.In contrast,CCRM1-1(-208 to-101)was critical for both expression specificity and level of Glu-1.Our findings not only provide new insights to uncover Glu-1 transcription regulatory machinery but also lay foundations for modifying Glu-1 expression. 展开更多

花生AhFAD2基因抑制表达的转基因后代分析

FAD2(Δ^12 fatty acid desaturase,Δ^12 FAD或FAD2)是催化油酸在脂肪酸碳链Δ^12位脱氢生成亚油酸的关键酶。在花生中,FAD2酶活性下降或失活可提高籽粒中油酸的相对含量,改善花生籽粒及制品的品质和氧化稳定性。通过将种子特异性表达Lectin启动子和Ca MV35S启动子驱动的倒位重复Ah FAD2基因RNAi干扰结构转入花生,获得了以丰花1号(FH1)和花育23(HY23)为受体、携带上述2种转化结构、稳定遗传的花生转基因纯合体株系,转基因花生主要农艺性状与非转基因对照基本一致。实时荧光定量分析发现,各转基因株系发育种子中Ah FAD2基因的转录水平普遍下调。气相色谱法进一步测定了部分转基因后代株系种子的脂肪酸含量及组成,籽粒中油酸含量分别平均提高了15.09%(HY23为受体)、36.40%(FH1为受体),相应地,亚油酸含量平均下降了16.19%、29.81%,油亚比平均增加了38.02%、98.10%。各转基因株系的油酸含量显著提高;且在以FH1为受体的转基因株系后代籽粒以及种子特异性启动子驱动的转化结构中,RNAi的抑制效果更明显。通过RNAi技术抑制花生FAD2基因的表达,可以有效提高花生籽粒油酸含量。该技术体系可以为花生品质育种提供借鉴。 展开更多
