OBJECTIVE:To investigate the protective effect and molecular mechanisms of Weining granule on N-methyl-N'-nitro-N-nitrosoguanidine (MNNG)-induced gastric cancer in rats.METHODS:A total of sixty healthy male wistar rats were randomly divided into five groups,including control group (CG),gastric cancer model group (MG),low-dose Weining granule treated group (LWT),medium-dose Weining granule treated group (MWT),and high-dose Weining granule treated group (HWT).Except the control group,the other groups were treated with MNNG to establish a rat model of gastric cancer.Low-dose Weining granule treated group,medium-dose Weining granule treated group,and high-dose Weining granule treated group were fed 9.0,18.0 and 36.0 g/kg Weining granule,respectively.Histopathologic and molecular biologic technology were adopted to determine the protective effect of Weining granule on MNNG-induced gastric cancer in rats.The pathological changes of gastrointestinal tissue were observed.Meanwhile,the differential expression of proliferation,apoptosis and angiogenesis markers were determined,including proliferating cell nuclear antigen (PCNA),pokemon,cyclin D1,B-cell lymphoma-2 (Bcl-2),caspase-3,phosphatase and tensin homolog (PTEN) and vascular endothelial growth factor (VEGF).RESULTS.:After the MNNG treated,the pathological changes of stomach tissue were improved noticeably,including the intestinal metaplasia and atypic hyperplasia.The experiment was completed in.58 rats (96.67%).As compared with gastric cancer model group,the general states of rats were improved significantly after treated with different dose Weining granule.Moreover,treatment with different doses of Weining granule could inhibit the protein and mRNA expression of PCNA,pokemon,cyclin D1,Bcl-2,and VEGF,while increase caspase-3 and PTEN (P < 0.01).CONCLUSION:Weining granule could improve gastric cancer by suppressing cell proliferation,promoting tumor cell apoptosis,and inhibiting angiogenesis. 展开更多

Some basic properties of granules,including the granule size distribution,packed-bed permeability,and chemical composition of the adhering layer,were investigated in this study for four iron ore blends consisting of 5wt%,25wt%,and 45wt%ultrafine magnetite and 25wt%ultrafine hematite concentrates.The effects of varying the sinter basicity(CaO/SiO2 mass ratio=1.4 to 2.2)and adding ultrafine concentrates on the variation of the adhering-layer composition and granule microstructure were studied.Moreover,the effect of adhering-layer compositional changes on sintering reactions was discussed in combination with pot sintering results of ore blends.Increasing sinter basicity led to an increase in the basicities of both the adhering layer and the fine part of the sinter mix,which were higher than the overall sinter basicity.When the sinter chemistry was fixed and fine Si-bearing materials(e.g.,quartz sand)were used,increasing the amount of ultrafine ores in the ore blends tended to reduce the adhering-layer basicity and increase the SiO2 content in both the adhering layer and the fine part of the sinter mix,which will induce the formation of low-strength bonding phases and the deterioration of sinter strength.The adhering-layer composition in granules can be estimated in advance from the compositions of the-1 mm fractions of the raw materials. 展开更多

为了进一步降低烟梗中的含丝率,课题组根据烟丝和烟梗的密度不同,设计了悬浮分离腔体结构。首先利用流体力学中的相关公式求解出通入悬浮分离腔体内的气压范围;再利用FLUENT软件对腔体内部的气流速度及轨迹进行模拟仿真,对比得到0.22MP... 为了进一步降低烟梗中的含丝率,课题组根据烟丝和烟梗的密度不同,设计了悬浮分离腔体结构。首先利用流体力学中的相关公式求解出通入悬浮分离腔体内的气压范围;再利用FLUENT软件对腔体内部的气流速度及轨迹进行模拟仿真,对比得到0.22MPa下的负压可以达到相对理想的分离效果;最后采用离散相模型和Euler-Lagrange多相流求解的方法,在FLUENT软件中对悬浮分离腔体内烟梗的颗粒相运动进行数值模拟仿真,得到烟丝颗粒与烟梗颗粒在腔体中的捕捉率。分离后的烟梗中的含丝率由原先的10%~20%降低到3.85%~8.53%,进一步提高了烟丝利用率。 展开更多

Isoamylase 1 (ISA1) is an isoamylase-type debranching enzyme which plays a predominant role in amylopectin synthesis. In this study, the clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated endonuclease 9 (CRISPR/Cas9) system was used to edit ISA1 gene in rice via Agrobacterium-mediated transformation. We identified 36 genetic edited lines from 55 T0 transgenic events, and classified the mutation forms into 7 types. Of those, two homozygous mutants, cr-isa1-1 (type 1, with an adenine insertion) and cr-isa1-2 (type 3, with a cytosine deletion) were selected for further analysis. Seed sizes of both cr-isa1-1 and cr-isa1-2 were affected, and the two mutants also displayed a shrunken endosperm with significantly lower grain weight. Electron microscopy analysis showed that abnormal starch granules and amyloplasts were found in cr-isa1-1 and cr-isa1-2 endosperm cells. The contents of total starch, amylose and amylopectin in the endosperm of the cr-isa1 mutants were significantly reduced, whereas sugar content and starch gel consistency were observably increased compared to the wild-type. The gelatinization temperature and starch chain length distributions of the cr-isa1 mutants were also altered. Moreover, transcript levels of most starch synthesis-related genes were significantly lower in cr-isa1 mutants. In conclusion, the results indicated that gene edition of ISA1 affected starch synthesis and endosperm development, and brought potential implications for rice quality breeding. 展开更多

Genomewide DNA Methylation Responses in Patients with β-Thalassemia Treated with Yisui Shengxue Granules (益髓生血颗粒)

Objective: To examine the clinical effects of Yisui Shengxue Granules(益髓生血颗粒) in the treatment of β-thalassemia and explore its mechanism on DNA methylation levels. Methods: A randomized placebo-controlled double-blinded trial was conducted. Forty patients with β-thalassemia were recruited and distributed randomly by envelope method into an experimental group and a control group, 20 patients in each group. The patients were given Yisui Shengxue Granules in the experimental group and placebo in the control group(12 g/bag, 3 times a day) during a 3-month intervention. Before and after 1, 2, and 3 months of treatment, peripheral intravenous blood was sampled, and blood parameters such as hemoglobin(Hb), red blood cells(RBCs), reticulocytes(Ret), and fetal hemoglobin(HbF) were analyzed. Mononuclear cells from 5 patients, who showed an obvious treatment effect, were isolated by density gradient centrifugation. DNA methylation was analyzed using an Affymetrix USA GeneChip Human Promoter 1.0 Array and Input-promoter 1.0. Results: Compared with pre-treatment, there was an obvious increase in Hb and RBCs counts after 1, 2, and 3 months in the experiment group(P<0.01 or P<0.05). Meanwhile, HbF increased from the 2 nd to the 3 rd month(P<0.05). In the control group, Hb and RBCs showed no obvioas change. After 3-month treatment, DNA methylation results from 5 patients revealed that there were 24 hypomethylated genes and 3,685 hypermethylated genes compared with pre-treatment. Genes of insulin-like growth factor 1 receptor(IGF1 R) and Janus kinase 3(JAK3) revealed the most relations with other genes(degree: 21) and genes of 1-phosphatidylinositol-4, 5-bisphosphate phosphodiesterase gamma 2(PLCG2) and mitogen-activated protein kinase 10(MAPK10) showed a stronger intermediary role(betweenness centrality=0.04). Conclusions: JAK3 and MAPK10 are two key genes in bone marrow and the lymphatic system, and JAK3 is likely to be related to hematopoietic cytokines in the process of early hematopoiesis.(Registration No. NCT 展开更多

Distinct functions of Trio GEF domains in axon outgrowth of cerebellar granule neurons

As a critical guanine nucleotide exchange factor(GEF) regulating neurite outgrowth, Trio coordinates multiple processes of cytoskeletal dynamics through activating Rac1, Cdc42 and RhoA small GTPases by two GEF domains, but the in vivo roles of these GEF domains and corresponding downstream effectors have not been determined yet. We established multiple lines of knockout mice and assessed the respective roles of Trio GEF domains and Rac1 in axon outgrowth. Knockout of total Trio in cerebellar granule neurons(CGNs) led to an impaired F-actin rearrangement of growth cone and hence a retarded neurite outgrowth. Such a retardation was reproduced by inhibition of GEF1 domain or knockdown of Cdc42 and restored apparently by introduction of active Cdc42. As Rac1 deficiency did not affect the neurite outgrowth of CGNs, we suggested that Trio GEF1-mediated Cdc42 activation was required for neurite outgrowth. We established a GEF2-knockout line with deletion of all Trio isoforms except a cerebella-specific Trio8, a short isoform of Trio without GEF2 domain, and used this line as a GEF2-deficient animal model. The GEF2-deficient CGNs had a normal neurite outgrowth but abolished Netrin-1-promoted growth, without affecting Netrin-1 induced Rac1 activation. We thus suggested that Trio GEF1-mediated Cdc42 activation rather than Rac1 activation drives the F-actin dynamics necessary for neurite outgrowth, while GEF2 functions in Netrin-1-promoted neurite elongation. Our results delineated the distinct roles of Trio GEF domains in neurite outgrowth, which is instructive to understand the pathogenesis of clinical Trio-related neurodevelopmental disorders. 展开更多

Methods: Relevant RCTs were searched through March 2019. Data were analyzed by Stata 15.0. Results: Nine articles involving 726 patients were enrolled in this study. Meta-analysis showed that the total effective rate [OR = 3.68 (2.44, 5.55), P < 0.001], Scr [SMD =-2.34 (-3.49,-1.19), P < 0.001], BUN [SMD =-1.80 (-2.73,-0.87), P < 0.001], Ccr [SMD = 0.71 (0.44, 0.97), P < 0.001] were better in the experimental group. But there were no significant difference in UA, CysC, 24h-Upro and incidence of adverse reactions (all P > 0.05) between two groups. No serious adverse reactions were found. Conclusions: The effect of the integrated medicine on CRF was better than Alprostadil alone. Uremic clearance granule is safe and has no obvious adverse reactions. 展开更多

传统的颗粒饲料淀粉含量检测方法具有准确度低、破坏样本等弊端。因此,文章研究了基于智能图像颗粒饲料淀粉含量检测方法,采用近红外光谱检测玉米籽粒内直链淀粉含量,分析玉米籽粒淀粉含量检测的特征波长优选方法,在不同频波段区间中,... 传统的颗粒饲料淀粉含量检测方法具有准确度低、破坏样本等弊端。因此,文章研究了基于智能图像颗粒饲料淀粉含量检测方法,采用近红外光谱检测玉米籽粒内直链淀粉含量,分析玉米籽粒淀粉含量检测的特征波长优选方法,在不同频波段区间中,通过相关系数法以及遗传算法优选近红外光谱建模变量,对玉米籽粒直链淀粉含量的定量分析模型进行构建,得到最优的淀粉含量波段为是波长(3 999~4 157+5 152~5 210 cm^-1),最佳PLS隐变量数是2个,最佳预处理方法是BASELINE+MC方法,最佳LS-SVM模型的RMSEP是0.027。将玉米透明度感官评分当成因变量,将图像透明度特征值当成自变量,通过逐步融入-过滤法实施多元线性回归分析,过滤出干扰玉米透明度的4个关键特征值(R值、B值、V值以及M值),塑造玉米透明度预测模型。通过双波长法检测玉米内直链以及支链淀粉含量和二者间的比例,研究透明度特征值同淀粉含量的相关性,以玉米直链淀粉含量为因变量,透明度特征值(M,h)为自变量,实施多元线性回归分析,构建玉米淀粉含量检测模型,实现玉米淀粉含量的准确检测。 展开更多

目的评价深、浅层脂肪室及不同大小颗粒的自体脂肪填充治疗面部容量缺失性老化的应用价值。方法2015年5月至2018年5月,四川华美紫馨医学美容医院美容外科用深、浅层脂肪室及自体脂肪颗粒移植技术用于面部容量缺失性老化女性患者126例,年... 目的评价深、浅层脂肪室及不同大小颗粒的自体脂肪填充治疗面部容量缺失性老化的应用价值。方法2015年5月至2018年5月,四川华美紫馨医学美容医院美容外科用深、浅层脂肪室及自体脂肪颗粒移植技术用于面部容量缺失性老化女性患者126例,年龄34~52岁,平均42.6岁。术后随访3个月至2.5年。应用视觉模拟评分法(VAS)对患者满意度进行评价。结果126例患者面部骨骼化外观较前圆润饱满,松垂减轻、皱纹减少,肤质改善,满意度(8.2±1.4)分。结论依据深、浅层脂肪室及不同大小脂肪颗粒填充能够恢复面部软组织容量,重塑面部轮廓和曲线,改善皮肤质地,是一种安全多效、满意度较高的面部容量缺失性老化的治疗方法。 展开更多

Alcohol, a widely abused drug, has deleterious effects on the immature nervous system. This study investigates the effect of chronic in vitro ethanol exposure on the metabolism of immature rat cerebellar granular cells (CGCs) and on their response to oxygen-glucose deprivation (OGD). Primary CGC cultures were exposed to ethanol (100 mM in culture medium) or to control ethanol-free medium starting day one in vitro (DIV1). At DIV8, the expression of ATP synthase gene ATP5g3 was quantified using real-time PCR, then cultures were exposed to 3 hours of OGD or normoxic conditions. Subsequently, cellular metabolism was assessed by a resazurin assay and by ATP level measurement. ATP5g3 expression was reduced by 12-fold (P = 0.03) and resazurin metabolism and ATP level were decreased to 74.4±4.6% and 55.5 ±6.9%, respectively after chronic ethanol treatment compared to control values (P<0.01). Additionally, after OGD exposure of ethanol-treated cultures, resazurin metabolism and ATP level were decreased to 12.7±1.0% and 9.0±2.0% from control values (P<0.01). These results suggest that chronic ethanol exposure reduces the cellular ATP level, possibly through a gene expression down-regulation mechanism, and increases the vulnerability to oxygen-glucose deprivation. Thus, interventions which improve metabolic function and sustain ATP-levels could attenuate ethanol-induced neuronal dysfunction and should be addressed in future studies. 展开更多

Background: Starch is the main source of energy in commonly used pig diets.Besides effects related to the extent of starch digestion,also several effects related to variation in digestion rate have recently been demonstrated in non-ruminants.Different rates of starch digestion in animals and in in vitro models have been reported,depending on the botanic origin of starch.Starches from different botanic sources differ widely in structural and molecular properties.Predicting the effect of starch properties on in vitro digestion kinetics based on existing literature is hampered by incomplete characterization of the starches,or by a selective choice of starches from a limited number of botanic sources.This research aimed to analyse the relationships between starch properties and in vitro digestion kinetics of pure starches isolated from a broad range of botanic origins,which are used in non-ruminant diets or have a potential to be used in the future.Therefore we studied starch digestion kinetics of potato,pea,corn,rice,barley,and wheat starches,and analysed the granule diameter,number of pores,type and amount of crystalline structure,amylose content and amylopectin side-chain length of all starches.Results: Multivariate analysis revealed strong correlations among starch properties,leading us to conclude that effects of most starch characteristics are strongly interrelated.Across all analysed botanic sources,crystalline type and amylopectin chain length showed the strongest correlation with in vitro digestion kinetics.Increased percentages of A–type crystalline structure and amylopectin side chains of DP 6–24 both increased the rate of digestion.In addition,within,but not across,(clusters of) botanic sources,a decrease in amylose content and increase in number of pores correlated positively with digestion kinetics.Conclusion: The type of crystalline structure and amylopectin chain length distribution of starch correlate significantly with digestion kinetics of starches across botanic sources in an in vitro pig mode 展开更多
