In vivo studies of animal models and in vitro studies of cell lines/primary... Investigating the pathophysiological mechanisms underlying brain disorders is a priority if novel therapeutic strategies are to be developed. In vivo studies of animal models and in vitro studies of cell lines/primary cell cultures may provide useful tools to study certain aspects of brain disorders. However, discrepancies among these studies or unsuccessful translation from animal/cell studies to human/clinical studies often occur, because these models generally represent only some symptoms of a neuropsychiatric disorder rather than the complete disorder. Human brain slice cultures from postmortem tissue or resected tissue from operations have shown that, in vitro, neurons and glia can stay alive for long periods of time, while their morphological and physiological characteristics, and their ability to respond to experimental manipulations are maintained. Human brain slices can thus provide a close representation of neuronal networks in vivo, be a valuable tool for investigation of the basis of neuropsychiatric disorders, and provide a platform for the evaluation of novel pharmacological treatments of human brain diseases.A brain bank needs to provide the necessary infrastructure to bring together donors, hospitals, and researchers who want to investigate human brain slices in cultures of clinically and neuropathologically well-documented material. 展开更多

目的分析男男性接触HIV感染者肛周尖锐湿疣的临床特征及微波联合光动力治疗的疗效。方法对260例使用微波联合光动力治疗肛周尖锐湿疣的男男性接触者的临床、实验室资料和疗效进行分析比较。结果肛周尖锐湿疣男男性接触者大多数为青年(24... 目的分析男男性接触HIV感染者肛周尖锐湿疣的临床特征及微波联合光动力治疗的疗效。方法对260例使用微波联合光动力治疗肛周尖锐湿疣的男男性接触者的临床、实验室资料和疗效进行分析比较。结果肛周尖锐湿疣男男性接触者大多数为青年(243/260),疣体位置以肛内型和肛内外型为主(245/260),其中HIV感染组肛外型所占比例(8/220)小于非HIV感染组(7/40)。所感染的HPV型别以11型(70/118)、6型(44/118)和16型(40/118)最常见,其中HIV感染者相比非HIV感染者更容易出现高危型、混合型和多重HPV感染。超过一半(146/260)的患者光动力治疗只需1~3次,且HIV感染组和非HIV感染组所需光动力治疗的次数和光敏剂的数量差异无统计学意义。HIV感染组复发率(13.6%,30/220)与非HIV感染组(15.0%,6/40)比较,差异无统计学意义,且远低于同期单纯行微波治疗的患者(45%,18/40)。结论微波联合光动力治疗能显著降低男男性接触HIV感染者肛周尖锐湿疣的复发率。 展开更多

Construction of a full-thickness human corneal substitute from anterior acellular porcine corneal matrix and human corneal cells

AIM: To construct functional human full-thickness corneal replacements.METHODS: Acellular porcine corneal matrix(APCM) was developed from porcine cornea by decellulariztion. The biomechanical properties of anterior-APCM(AAPCM) and posterior-APCM(PAPCM) were checked using uniaxial tensile testing. Human corneal cells were obtained by cell culture. Suspending ring was designed by deformation of an acupuncture needle. MTT cytotoxicity assay was used to check the cytotoxicity of suspending ring soaking solutions. A new three-dimensional organ culture system was established by combination of suspending ring, 48-well plate and medium together. A human full-thickness corneal substitute was constructed from human corneal cells with AAPCM in an organ coculture system. Biochemical marker expression of the construct was measured by immunofluorescent staining and morphological structures were observed using scanning electron microscopy. Pump function and biophysical properties were examined by penetrating keratoplasty and follow-up clinical observations.RESULTS: There were no cells in the AAPCM or PAPCM, whereas collagen fibers, Bowman’s membrane, and Descemet’s membrane were retained. The biomechanical property of AAPCM was better than PAPCM. Human corneal cells grew better on the AAPCM than on the PAPCM.There was no cytotoxicity for the suspending ring soaking solutions. For the constructed full-depth human corneal replacements keratocytes scattered uniformly throughout the AAPCM and expressed vimentin. The epithelial layer was located on the surface of Bowman’s membrane and composed of three or four layers of epithelial cells expressing cytokeratin 3. One layer of endothelial cells covered the stromal surface of AAPCM, expressed Na+/K+ATPase and formed the endothelial layer. The construct was similar to normal human corneas, with many microvilli on the epithelial cell surface, stromal cells with a long shuttle shape, and zonula occludens on the interface of endothelial cells. BACKGROUND Femoral neck stress fractures are rarely encountered among young adults and are often associated with either repetitive excessive loading or underlying bone pathology.Preliminary research has indicated human immunodeficiency virus(HIV)/antiretroviral therapy(ART)as predisposing agents to osteopenia and osteoporosis related complications.We report a case of HIV/ART induced insufficiency fracture in a resource limited setting in Central India.Our aim is to increase awareness and promote screening of HIV/ART related osteopenia and osteoporosis in order to prevent catastrophic orthopaedic complications.CASE SUMMARY A 35-year-old HIV positive male presented with a stress fracture of left femoral neck.The patient was on ART and reported no comorbidities.He went on to be successfully managed surgically.However,during work-up osteopenia of the contralateral proximal femur was recognised using Singh’s Index.Six months post-op the patient presented with right-sided femoral-neck stress fracture.At this stage the patient was nonconcordant with ART and denied surgical fixation.CONCLUSION In the absence of co-morbidities,several mechanisms of HIV/antiretroviral therapy may have played a role in predisposing our patient towards such a presentation.We recommend routine screening all HIV-infected patients for osteopenia,especially in younger individuals.In low resource settings and district hospitals,pelvis radiograph&Singh’s index can be used for screening. 展开更多

Eosinophils are currently regarded as versatile mobile cells controlling and regulating multiple biological pathways and responses in health and disease.These cells store in their specific granules numerous biologically active substances(cytotoxic cationic proteins, cytokines, growth factors, chemokines,enzymes) ready for rapid release. The human gut is the main destination of eosinophils that are produced and matured in the bone marrow and then transferred to target tissues through the circulation. In health the most important functions of gut-residing eosinophils comprise their participation in the maintenance of the protective mucosal barrier and interactions with other immune cells in providing immunity to microbiota of the gut lumen. Eosinophils are closely involved in the development of inflammatory bowel disease(IBD),when their cytotoxic granule proteins cause damage to host tissues. However,their roles in Crohn’s disease and ulcerative colitis appear to follow different immune response patterns. Eosinophils in IBD are especially important in altering the structure and protective functions of the mucosal barrier and modulating massive neutrophil influx to the lamina propria followed by transepithelial migration to colorectal mucus. IBD-associated inflammatory process involving eosinophils then appears to expand to the mucus overlaying the internal gut surface. The author hypothesises that immune responses within colorectal mucus as well as ETosis exerted by both neutrophils and eosinophils on the both sides of the colonic epithelial barrier act as additional pathogenetic factors in IBD. Literature analysis also shows an association between elevated eosinophil levels and better colorectal cancer(CRC) prognosis, but mechanisms behind this effect remain to be elucidated. In conclusion, the author emphasises the importance of investigating colorectal mucus in IBD and CRC patients as a previously unexplored milieu of disease-related inflammatory responses. 展开更多

Resorting to the immensely state-centric international legal system to regulate corporate human rights abuses is often viewed as inadequate.Among many proposals aiming at filling the international regulatory gaps,imposing international human rights obligations directly on corporations is a bold one,which,due to profound doctrinal and practical challenges,is yet to be materialized.However,state-owned enterprises(SOEs),given their prima facie''state-business nexus"that blurs the traditional public-private divide,might provide a renewed opportunity to push forward the"direct international corporate accountability^^campaign.This study investigates whether SOEs represent a golden chance for direct corporate accountability in the international legal regime.This study provides a legal analysis supported by case law,and by comparative and empirical research when appropriate.After providing a definitional account of SOEs,it examines the legal status of SOEs under international law.Then,in the reverse direction,it proceeds to explore if the state-business nexus of SOEs as non-state actors could render the argument toward direct international corporation accountability more convincing.Major findings reveal that SOEs,to a limited extent,represent a renewed opportunity to rethink direct corporate accountability under international law. 展开更多

Currently,there does not exist a strategy that can reduce diabetes and scientists are working towards a cure and innovative approaches by employing stem cellbased therapies.On the other hand,bioprinting technology is a novel therapeutic approach that aims to replace the diseased or lostβ-cells,insulin-secreting cells in the pancreas,which can potentially regenerate damaged organs such as the pancreas.Stem cells have the ability to differentiate into various cell lines including insulinproducing cells.However,there are still barriers that hamper the successful differentiation of stem cells intoβ-cells.In this review,we focus on the potential applications of stem cell research and bioprinting that may be targeted towards replacing theβ-cells in the pancreas and may offer approaches towards treatment of diabetes.This review emphasizes on the applicability of employing both stem cells and other cells in 3D bioprinting to generate substitutes for diseasedβ-cells and recover lost pancreatic functions.The article then proceeds to discuss the overall research done in the field of stem cell-based bioprinting and provides future directions for improving the same for potential applications in diabetic research. 展开更多

Here we reported a Chinese case of bilateral peripheral facial paralysis (PFP) in human immunodeficiency virusc (HIV) infected population. A 38-year-old homosexual male patient was referred to our hospital for bilateral facial paralysis. 21 days prior to admission he had developed high fever, chills, headache, fatigue, general malaise, nausea and vomiting. Neurological examination revealed bilateral ptosis of lower lip and cheeks, as well as failure of bilateral eyes closure. Analysis of cerebrospinal fluid (CSF) revealed pleocytosis, a marked rise of micro total protein and a marked rise of intrathecal lgG synthesis. The result of HIV-1 serology was positive by ELISA and that was confirmed by western blot. His CD4+ cell count was 180 cells/mm3. HIV-1 viral load in CSF was almost 10 times higher than that in plasma. The patient’s condition improved steadily and experienced complete resolution of bilateral PFP after 2 months. 展开更多

This paper proposes a strategy to analyzeand predict human error on the basis of a further modification of theexisting cognitive reliability and error analysis method (CREAM)in HRA. Through providing a broader definition for the key parametersused in the quantification method, this paper produces arelatively flexible strategy to determine the nominal HEP (HEP0).Basing on this strategy, the actual quantification of HEP in CREAMis able to be applied to more verified conditions. To prove the validityof the method proposed, a case study of spacecraft launch isalso introduced, in which the prediction derived from the method isproved consistent with real field data. 展开更多

Olanzapine (OLP), chemically known as 2-Methyl-10-(4-methyl-piperazin-1-yl)-4H-3-thia-4,9-diaza-benzo[f]azulene, is an atypical antipsychotic drug. It is used for the treatment of schizophrenia and bipolar disorder. A new simple and selective membrane based potentiometric sensor was developed for potentiometric determination of olanzapine. The membrane was constructed using an ion-pair of OLP and sodium tetraphenyl boron in dioctyl phthalate and PVC. The membrane provides good linear Nernstian response covering relatively wide concentration range of 4 × 10^-6 - 1 × 10^-2 M OLP over pH range of 2.6-7.8. The detection limit for the developed sensor was founded as 2.02 × 10^-6 M. The response time of developed sensor is <10 s for the range of determination. The sensor showed good selectivity for OLP in the presence of various cations, anions and other organic molecules. The membrane was successfully applied in direct potentiometric determination of OLP in tablets. The percentage recovery of OLP, ranged from 96.2 to 99.68% with a mean standard deviation <5% indicates the adoptability of sensor for the direct estimation of OLP in pharmaceuticals. The developed sensor was used to determine OLP in spiked human urine sample and the satisfactory results were obtained. 展开更多

On the other hand, with the increasing degree of interdependence, the threats and challenges facing all countries worldwide are becoming more common. In particular, issues such as climate change, financial crises, large-scale outbreak of infectious diseases, and cyberattacks, have become globalized, and need to be addressed through global governance. Phenolic Profile, Antioxidant Activity and Anti-proliferative Activity of Crabapple Fruits

To explore the potential of crabapples as functional food, polyphenols in crabapples and ‘Fuji’ apples were extracted, and the phenolic profile,total polyphenols, antioxidant activity and anti-proliferative activity against several human cancer cells were determined. The results indicated that crabapple extracts have more abundant phenols and higher total polyphenols(from 4.46 to 46.63 mg GAE·g-1 DW) compared to ‘Fuji’ apples.Crabapple extracts possessed higher antioxidant activity than apple by DPPH and ABTS analysis. All fruit extracts exhibited inhibitory effects on proliferation in different cancer cells;however, crabapple extracts performed significantly better, with half inhibitory concentration(IC50)values varied from 48.34 μg·m L-1 to 974.81 μg·m L-1 for colon cancer cells SW480, 64.67–1 466.35 μg·m L-1 for stomach cancer cells BGC-803,78.88–910.64 μg·m L-1 for esophageal cancer cells CaEs-17. Besides, the red crabapples had higher antioxidant activity and anti-proliferative activity than yellow fruits. These results showed that crabapples, especially red crabapples, have great potential as a healthy food, as they are rich in phenolic compounds with high antioxidant and anti-proliferative activities to cancer cells. 展开更多

Neural stem cells,which are capable of multi-potential differentiation and self-renewal,have recently been shown to have clinical potential for repairing central nervous system tissue damage.However,the theme trends and knowledge structures for human neural stem cells have not yet been studied bibliometrically.In this study,we retrieved 2742 articles from the PubMed database from 2013 to 2018 using "Neural Stem Cells" as the retrieval word.Co-word analysis was conducted to statistically quantify the characteristics and popular themes of human neural stem cell-related studies.Bibliographic data matrices were generated with the Bibliographic Item Co-Occurrence Matrix Builder.We identified 78 high-frequency Medical Subject Heading(MeSH)terms.A visual matrix was built with the repeated bisection method in gCLUTO software.A social network analysis network was generated with Ucinet 6.0 software and GraphPad Prism 5 software.The analyses demonstrated that in the 6-year period,hot topics were clustered into five categories.As suggested by the constructed strategic diagram,studies related to cytology and physiology were well-developed,whereas those related to neural stem cell applications,tissue engineering,metabolism and cell signaling,and neural stem cell pathology and virology remained immature.Neural stem cell therapy for stroke and Parkinson’s disease,the genetics of microRNAs and brain neoplasms,as well as neuroprotective agents,Zika virus,Notch receptor,neural crest and embryonic stem cells were identified as emerging hot spots.These undeveloped themes and popular topics are potential points of focus for new studies on human neural stem cells. 展开更多

DNA methyltransferase 1(DNMT1)is a useful biomarker for lung cancer in early clinical diagnosis.A rapid magnetic chemiluminescence immunoassay(MCLIA)for DNMT1 in human serum has been developed.Horseradish peroxidase(HRP)-second-Ab was used to labeled polyclonal antibodies of anti-DNMT1.DNMT1 in sample integrates with specific immunomagnetic beads and can constitute a supersandwiched immunoreaction.In magnetic field,nonspecific materials can be separated.After luminescent substrate luminol-H2O2-BIP was added,the relative light unit(RLU)of HRP was detected and was discovered to be directly proportional to the content of DNMT1 in sample.The correlative variables involved in the MCLIA value were optimized and the methodological evaluation was carried out.After optimization,in the range of0.5–128 ng/mL,the linear regression equation was y=0.5014 x+1.769(x was logCDNMT1,y was relative luminescence units(RLU)/RLU0),and the limit of detection was 0.01 ng/mL.The RSD of intra-and interassays were 15.8%–16.9%and 14.3%–18.1%,respectively.The recovery was from 70.0%to 106.2%.Furthermore,paralleled with purchasable enzyme-linked immunosorbent assay(ELISA)kits,MCLEIA had lower detection limit,wider linear range and shorter detection time.Therefore,the MCLEIA established in this study could be used for the sensitive detection of DNMT1 in serum sample. 展开更多

Understanding urban dynamics and large-scale human mobility will play a vital role in building smart cities and sustainable urbanization. Existing research in this domain mainly focuses on a single data source (e.g., GPS data, CDR data, etc.). In this study, we collect big and heterogeneous data and aim to investigate and discover the relationship between spatiotemporal topics found in geo-tagged tweets and GPS traces from smartphones. We employ Latent Dirichlet Allocation-based topic modeling on geo-tagged tweets to extract and classify the topics. Then the extracted topics from tweets and temporal population distribution from GPS traces are jointly used to model urban dynamics and human crowd flow. The experimental results and validations demonstrate the efficiency of our approach and suggest that the fusion of cross-domain data for urban dynamics modeling is more practical than previously thought. 展开更多

Objective:To investigate the relationship between body mass index(BMI),waist circumference(WC),hip circumference(HC),waist-hip ratio(WHR),waist height ratio(WHtR),fasting blood glucose(FPG),fasting blood glucose variation(FPG-CV)and glycosylated hemoglobin(HbA1C)to provide a reference for predicting the risk and development trend of diabetes mellitus.Methods:From October 2016 to December 2017,111 subjects from the Second Affiliated Hospital of Tianjin University of Traditional Chinese Medicine were selected to measure their height and weight.BMI,WHR and WHtR were calculated and the concentrations of FPG and HbA1C were detected.Statistical analysis was carried out with SPSS21.0 software,and the data were processed by multiple linear regression analysis.Results:The concentration dependence of WHtR,FPG,FPG-CV and HbA1C were more closely related.There was a significant difference between WHtR and FPG,FPG-CV and HbA1C by multiple linear regression analysis(t=8.531,6.910 and 6.905,respectively,P<0.01),and the correlation coefficient was 0.633,0.552 and 0.552 respectively(P<0.01).Conclusion:There is a significant correlation between WHtR and FPG,FPG-CV and HbA1C.Therefore,measuring the height and waist circumference for the predictive potential of diabetes needs to be emphasized and intensified. 展开更多
