Abrupt visual loss during anti-vascular endothelial growth factor treatment for type 3 neovascularization

AIM: To investigate the incidence of abrupt visual loss and its associated factors, during anti-vascular endothelial growth factor(VEGF) treatment for type 3 neovascularization.METHODS: This retrospective study included 137 eyes that were newly diagnosed with type 3 neovascularization. All eyes were treated with anti-VEGF therapy. Abrupt visual loss was defined as loss of 5 or more lines in best-corrected visual acuity(BCVA) in comparison to the previous visit. The incidence and timing of abrupt visual loss as well as the factors associated with it, were determined. In addition, the BCVA at the final follow-up was compared between the eyes with and those without abrupt visual loss.RESULTS: The mean follow-up period was 42.4±18.9 mo after diagnosis, and abrupt visual loss was noted in 22 eyes(16.1%) at a mean of 19.6±13.9 mo. Abrupt visual loss was found to be associated with subretinal hemorrhage in 11 eyes(50.0%), development of or increase in the height of pigment epithelial detachment with fluid in 8 eyes(36.4%), and tears in the retinal pigment epithelium in 3 eyes(13.6%). The logarithm of minimum angle of resolution(logMAR) mean BCVA at the final follow-up was 2.07±0.67(Snellen equivalents: 20/2349) and 1.00±0.55(20/200) in eyes with and without abrupt visual loss, respectively. BCVA was significantly worse in the eyes with abrupt visual loss(P<0.001).CONCLUSION: Abrupt visual loss is noted in 16.1% of patients with type 3 neovascularization and is associated with poor visual outcome. Additional studies are needed to determine how abrupt visual loss can be prevented. 展开更多

AIM: To investigate the regulation of vascular endothelial growth factors (VEGF) and pigment epithelium-derived factor (PEDF) expression by autophagy in retinal pigment epithelium (RPE) cells on exposure to hypoxia. METHODS: ARPE-19, an RPE cell line, was treated as following: the control group was kept in a normoxic incubator;the hypoxia group was incubated in a hypoxic incubator with 1% O2/5% CO2/94% N2 for 24h;the hypoxia + 3-methyladenine (3-MA) group was pretreated with 10 mmol/L 3-MA for 1h and then in the hypoxic incubator for 24h;and the hypoxia + chloroquine (CQ) group was pretreated with 50 μmol/L CQ for 1h and then in the hypoxic incubator for 24h. The morphology and ultrastructure of the cells was observed by an inverted microscope or a transmission electronic microscope (TEM). Western blot was performed to assay the expression of autophagy-associated markers, including microtubule associated protein 1 light chain 3 B (LC3B), Beclin-1, Atg5 and p62. The concentration of VEGF and PEDF in the culture supernatant was determined by ELISA, and the ratio of VEGF/PEDF was calculated. RESULTS: There were no obvious differences in cell morphology among different groups and autolysosomes could be observed in the cytoplasm in all groups. Compared to the control cells, the LC3B-II/I ratio and levels of Beclin-1 and Atg5 were significantly increased and p62 level was significantly decreased in the hypoxia group. With the increase of VEGF and decrease of PEDF concentration, the VEGF/PEDF ratio was significantly increased in the hypoxia group compared to the control cells. The LC3B-II/I ratio was significantly reduced by 3-MA treatment and increased by CQ treatment. The expressions of Beclin-1 and Atg5 were significantly reduced by 3-MA or CQ treatment, while expression of p62 was increased in the 3-MA or CQ treated cells. AIM: To evaluate the expression of uncoupling protein 2(UCP2) in a retinal pigment epithelium cell line(ARPE-19), under oxidative stress(OS).METHODS: ARPE-19 cells were divided into groups treated with various concentrations of hydrogen peroxide(H2 O2;0, 150, 300, 500, 700, and 900 μmol/L) for 24 h, to induce oxidative damage and cell viability was assessed by MTT assay. UCP2 mRNA expression in cells treated with H2 O2 was investigated by reverse transcription-polymerase chain reaction(RT-PCR). UCP2 protein expression was assessed by Western blotting and ROS levels analyzed by flow cytometry(FCM). Further, UCP2-siRNA treated cultures were exposed to H2 O2(0, 75, 150, and 300 μmol/L) for 2 h and cell viability determined by MTT assay.RESULTS: Cells treated with higher concentrations of H2 O2 appeared shrunken;their adhesion to adjacent cells was disrupted, and the number of dead cells increased. The results of cell viability assays demonstrated that the numbers of cells were decreased in a dose-dependent manner following treatment with H2 O2. Compared with untreated controls, cell viability was significantly reduced after treatment with >300 μmol/L H2 O2(P<0.05). Cell metabolic activity was decreased with increased concentrations of H2 O2 as detected by MTT assay. Levels of OS were further decreased in cells treated with UCP2-siRNA compared with those treated with H2 O2 alone(P<0.05). The results of RT-PCR and Western blotting demonstrated that UCP2 expression was reduced in H2 O2-treated groups compared with controls(P<0.05). FCM analysis showed that cell reactive oxygen species(ROS) levels were increased in H2 O2-treated groups and further upregulated by UCP2-si RNA treatment(P<0.05).CONCLUSION: Expression levels of UCP2 are decreased in ARPE-19 cells treated with H2 O2. ROS levels are further increased in cells treated with UCP2-siRNA relative to those treated with H2 O2 alone. UCP2 may have a protective role in ARPE-19 cells during oxidative injury. 展开更多

本研究对实验室购买、收集的9株红曲菌株分泌麦角固醇、色素和桔霉素的能力进行比较分析,并探讨了三者的相关性,旨在为红曲菌资源的深度挖掘提供基础信息。分别采用UPLC-紫外检测法、UV-VIS法和UPLC-荧光检测法测定9株红曲菌发酵制备红... 本研究对实验室购买、收集的9株红曲菌株分泌麦角固醇、色素和桔霉素的能力进行比较分析,并探讨了三者的相关性,旨在为红曲菌资源的深度挖掘提供基础信息。分别采用UPLC-紫外检测法、UV-VIS法和UPLC-荧光检测法测定9株红曲菌发酵制备红曲样品中麦角固醇、色价和桔霉素的含量;在此基础上,采用统计学分析方法分析了三者之间的相关性。分析表明,不同菌株产麦角固醇、色素和桔霉素的能力不同,其中紫色红曲菌OUT 2011发酵红曲中麦角固醇含量和色价均最高,分别达到(1121.96±55.06) mg/kg和(1058.97±5.04) U,安卡红曲菌AS 3.554发酵红曲中麦角固醇含量和色价均最低,而从毛红曲菌OUT 2137、红色红曲菌AS 3.549,安卡红曲菌AS 3.554、发白红曲菌CICC5017、橙色红曲菌CICC5015发酵的红曲均未检出桔霉素。9株红曲菌发酵产麦角固醇、色素和桔霉素能力各不相同;相关性分析显示菌株分泌的麦角固醇和色素水平呈正相关性,为筛选高产麦角固醇的红曲菌株提供了参考。 展开更多

The role of heat shock protein 70 (HSP70) in apoptosis of human retinal pigment epithelial cells (ARPE-19) induced by 4-hydroxy-2-nonenal (4-HNE) was explored. Different concentrations of 4-HNE were used to stimulate ARPE-19 cells, and apoptosis was measured by flow cytometry. The expression of apoptotic-related proteins, HSP70, X-linked inhibitorof- apoptosis (XIAP), Bcl-2, and Bax were quantified by Western blotting. HSP70 and XIAP overexpression plasmids, or their corresponding siRNAs were transfected into ARPE-19 cells using Lipofectamine. 2000. Co-immunoprecipitation and Western blotting were used to detect the effect of 4-HNE on the expression of HSP70 and the binding level between 4-HNE and HSP70. The results showed that 4-HNE induced late apoptosis in ARPE-19 cells, accompanied by elevated levels of 4-HNE-modified IISP70, but it did not affect HSP70 protein expression. 4-HNE-modified HSP70 down-regulated the expression of the apoptosis inhibitory protein XIAP. Overexpression of HSP70 or XIAP inhibited 4-HNE-induced apoptosis of ARPE-19 cells. It was suggested that 4-HNE could promote XIAP degradation by modification of HSP70 to induce late apoptosis of human retinal pigment epithelial cells. 展开更多

响应面法优化紫五加色素提取工艺及其稳定性研究

以紫五加为原料,利用超声波辅助法提取紫五加色素.在单因素实验的基础上,采用响应面分析法优化提取方案.结果表明,紫五加色素最佳提取工艺条件(修正)为:液料比为60∶1,乙醇体积分数为85%,超声功率320W,超声时间94min.此工艺条件下提取... 以紫五加为原料,利用超声波辅助法提取紫五加色素.在单因素实验的基础上,采用响应面分析法优化提取方案.结果表明,紫五加色素最佳提取工艺条件(修正)为:液料比为60∶1,乙醇体积分数为85%,超声功率320W,超声时间94min.此工艺条件下提取的紫五加(烘干枝梢部分)色素吸光度值可达0.756(预测值0.760).各部分色素含量顺序:c(叶)>c(枝梢)>c(茎)>c(根).除叶外,前处理方式(烘干或风干)对紫五加色素提取影响不大.紫五加色素稳定性研究表明:抗氧化剂(异抗坏血酸,C6H8O6)、氧化剂(H2O2)、还原剂(Na2SO3)和蔗糖均明显影响紫五加色素稳定性;金属离子Ca^2+,Na^+和Fe^3+对紫五加色素的影响较小,而Mg^2+,Al^3+,Zn^2+,Cu^2+和Mn^2+影响较大;10℃~90℃温度范围内,溶液pH<7,自然光或紫外光照射,加入NaCl溶液均对紫五加色素无明显影响. 展开更多

AIM: To analyze the expression of uncoupling protein 2(UCP2) in retinal pigment epithelium(RPE) cells at the different human age, further explore the possible new target of RPE cells protection.METHODS: Adult retinal pigment epithelial-19(ARPE-19) cells and the primary RPE cells at the different age(9-20 y,50-55 y, 60-70 y, >70 y) were cultured and harvested. The expression of UCP2 in these cells was detected by reverse transcription-polymerase chain reaction(RT-PCR), Western blot and confocal microscopy.RESULTS: Cells from the donors more than 60 y are larger and more fibroblastic in appearance compared to ARPE-19 cells and those primary cultures obtained from the younger individuals by using phase-contrast micrographs. Results of RT-PCR, Western blot and confocal microscopy all showed that UCP2 was highly expressed in ARPE-19 cells and in the younger primary cultured human RPE cells at the age of 9-20 y and 50-55 y, whereas lower expression of UCP2 was measured in the older primary cultured human RPE cells at the age more than 60 y.CONCLUSION: Expression of UCP2 gene is decreased in aged RPE cells, promoting the lower ability of anti-oxidation in these cells. 番茄的颜色和色素含量是决定果实品质的两个重要特性。在本研究中,使用绿色和紫色番茄植株分别作为母本和父本,分别产生了6个遗传群体,即P1、P2、F1、BC1、BC2和F2。通过数量性状的多代联合分析研究果实颜色和色素含量的遗传规律。结果... 番茄的颜色和色素含量是决定果实品质的两个重要特性。在本研究中,使用绿色和紫色番茄植株分别作为母本和父本,分别产生了6个遗传群体,即P1、P2、F1、BC1、BC2和F2。通过数量性状的多代联合分析研究果实颜色和色素含量的遗传规律。结果表明,绿色和紫色果之间的果实颜色的最佳遗传模型为MX1-AD-ADI。BC1、BC2和F2代色度值的主基因遗传率分别为82%、59%和77%,多基因遗传率分别为8%、10%和13%。番茄红素含量的遗传符合MX2-ADI-AD模型,BC1、BC2和F2的主基因遗传率分别为64%、74%和93%,而多基因的遗传率分别为20%、0%和0%。叶绿素的遗传模型为MX1-AD-ADI,BC1、BC2和F2的主基因遗传率分别为59%、74%和59%,多基因的遗传率均为0%。胡萝卜素的遗传符合MX2-ADI-ADI模型,BC1、BC2和F2的主基因遗传率分别为76%、4%/5%和54%,而多基因的遗传率分别为0%、0%和13%。 展开更多

为了探寻影响紫花槭叶色变化的主要因素,从而为其园林配置提供理论依据,对紫花槭秋季叶片转色不同时期的叶色参数、色素含量、相关酶活性、碳水化合物和矿质元素含量进行了测定。结果表明:1)紫花槭叶色参数L值先上升后降低,a值呈持续上... 为了探寻影响紫花槭叶色变化的主要因素,从而为其园林配置提供理论依据,对紫花槭秋季叶片转色不同时期的叶色参数、色素含量、相关酶活性、碳水化合物和矿质元素含量进行了测定。结果表明:1)紫花槭叶色参数L值先上升后降低,a值呈持续上升趋势,b值呈先上升后下降再上升的趋势;2)紫花槭叶片叶绿素含量呈下降趋势,类胡萝卜素含量呈先下降后上升再下降的趋势,花色素苷含量呈上升趋势;3)苯丙氨酸解氨酶(PAL)、查尔酮异构酶(CHI)、查尔酮还原酶(CHR)活性均呈先升高后降低的趋势;4)碳水化合物中蔗糖含量呈先下降后上升再下降的趋势;5)矿质元素中氮、钾含量的变化趋势相同,均呈下降趋势,磷含量呈先上升后下降的双峰趋势,镁含量呈先上升后下降的趋势,钙含量呈先下降后上升再下降的趋势。对不同色素与其他生理指标的相关性分析结果表明:叶绿素含量与氮、钾均存在极显著的正相关关系,与钙存在显著的正相关关系;类胡萝卜素含量与其他指标的相关性均未达到显著水平;花色素苷的含量与PAL、CHI酶活性和氮、钾含量间均呈极显著负相关,与CHR酶活性及钙、镁含量间均呈显著负相关,而与淀粉、蔗糖含量间均存在正相关,但不显著;色素与磷含量无明显的相关性。 展开更多

以烟用烤房内生产豌豆苗为材料,研究不同光质(蓝光、红光、蓝红混合光)对豌豆苗长、重量积累、色素含量及营养物质含量的影响。结果表明,相同光照条件下,相对较高的喷水频度能够提高豌豆苗长、鲜重和干重、色素含量和营养成分含量,其中... 以烟用烤房内生产豌豆苗为材料,研究不同光质(蓝光、红光、蓝红混合光)对豌豆苗长、重量积累、色素含量及营养物质含量的影响。结果表明,相同光照条件下,相对较高的喷水频度能够提高豌豆苗长、鲜重和干重、色素含量和营养成分含量,其中蓝光照射条件下,叶绿素a、VC含量和总酚含量显著增加,红光照射条件下,叶绿素b、类黄酮和总酚含量显著增加,蓝红混合光照条件下,叶绿素a、叶绿素b、类胡萝卜素、VC含量和类黄酮含量显著增加。相同喷水频度下,蓝光处理能够显著提升苗长及显著增加豌豆苗茎叶鲜重、总鲜重、总干重。不同光质对豌豆苗叶片中不同色素含量的影响存在差异,蓝光处理显著增加叶绿素a含量,红光处理显著增加叶绿素b含量,蓝红混合光照处理显著增加类胡萝卜素含量。红光处理酚类物质含量显著高于其他光质。蓝光照射处理VC含量、可溶性蛋白含量和类黄酮含量高于红光照射。豌豆苗中色素总量与VC含量、类黄酮含量呈极显著正相关,与可溶性蛋白含量呈显著正相关。总之,豌豆苗生产过程中采用蓝光照射配合较高的喷水频度能够改善豌豆苗生长发育状况和达到提升豌豆苗营养价值的目的。 展开更多
