Objective:To investigate the effect of radiotherapy plus recombinant human endostatin(RHendostatin) on esophageal cancer and its mechanism.Methods:A total of SO nudemice were equally randomized into control group,radiotherapy group,and combined therapy group Ⅰ,Ⅱ,and Ⅲ after inoculating with Ecal09 cell suspension(1×107 cells/mL).On the day of grouping,control group and radiotherapy group were injected normal saline,while radiotherapy group and 3 combined therapy groups received radiotherapy;besides,combined therapy group Ⅰ,Ⅱ,and Ⅲ was injected RH-endostatin of 2.5,5,10 mg/kg respectively.After 3-week therapy,the tumors of each group were collected and microvessel density and VEGF expression in tumors were determined.In vitro,Eca109 cells were divided into control group,radiotherapy group,and combined therapy group.Forty-eight hours after treatment cell cycle distribution and apoptosis rate were detected,and the activity of VEGF signal paths was semiquantitatively analyzed.Results:Since the 6th day of treatment,the relative tumor proliferation rate of combined therapy group Ⅱ was lower than radiotherapy group(P<0.05) and 40%since the 15 th day.Average microvessel density and EGFR expression in combined therapy group Ⅱ were lower than radiotherapy group(P<0.05).In vitro,the cell percentage in S and G2/M phase of combined therapy group cells was lower than that in radiotherapy group cells,while the apoptosis rate and the expression of VEGF,AKT,p-AKT,ERK1/2 and p-ERKl/2 in combined group were higher than that in radiotherapy group(P<0.05).Conclusions:RH-endostatin promotes the efficacy of radiotherapy on esophageal cancer,which may be partly realized by inhibiting the activity of VEGF related signal paths. 展开更多

AIM: To evaluate the safety and efficacy of submucosal tunneling and endoscopic resection(STER) for treating submucosal tumors(SMTs).METHODS: Between August 2012 and October 2013, 21 patients with SMTs originating from the muscularis propria(MP) layer at the esophagogastric junction were treated by STER of their tumors. Key steps of the procedure include:(1) mucosal incision: a 2-cm longitudinal mucosal incision was made 5 cm proximal to the tumor;(2) submucosal tunneling: a submucosal tunnel was created 5 cm proximal to and 1 to 2 cm distal to the tumor;(3) tumor resection: the SMT was resected under direct endoscopic viewing;(4) hemostasis: while finishing the tumor resection, careful hemostasis of the MP defect and the tunnel was performed; and(5) mucosal closure: the mucosal incision site was closed by using hemostatic clips. During the operation, equipment used included a cap-fitted endoscope, an insulatedtip knife, a hook knife, hemostatic forceps, an injection needle, a snare, an endoclip, and a high-frequency generator. Carbon dioxide(CO2) insufflation was achieved by using a CO2 insufflator.RESULTS: The median age of the patients was 46.2 years(range, 35-59 years), and the majority were male(18 male vs 3 female). Complete resection rate was 100%(21/21). Eighteen lesions were resected en bloc. Mean tumor size was 23 mm(range, 10-40 mm), and mean procedure time was 62.9 min(range, 45-90 min). Pathological diagnosis of these tumors included leiomyoma(15 out of 21) and gastrointestinal stromal tumor(6 out of 21). Full-thickness MP resection was performed in 9 of 21 patients(42.9%), with mediastinal and subcutaneous emphysema occurring in all nine. At the completion of the procedure, all patients received closure of the incision with hemoclips. One patient required percutaneous drainage. The remaining 20patients required no further endoscopic or surgical intervention. There were no incidents of massive or delayed bleeding. Based on the c DNA sequences from hyper variable(HV) regions of identified 52 S-alleles in Oriental pear cultivars, S-RNase c DNA probes were designed, and a c DNA microarray for S-RNase detections was established. Each microarray contained 240 sites from 55 c DNA probes, including all specific c DNA sequences from the HV regions of the S-alleles. Using the c DNA of pistils of tested pear cultivars as template and Cy3 fluorescently labeling primers by PCR amplification, microarray hybridization detected the S-genotype of each pear cultivar. The genotypes inferred from the c DNA microarray hybridization signals of pear cultivars such as ‘Lijiang Huangsuanli’, ‘Xiuyu’, ‘Midu Yuli’, ‘Baimianli’, and ‘Deshengxiang’ were similar to the known genotypes of all tested cultivars. The S-RNase c DNA microarrays and the oligonucleotide gene chips were then used to conduct parallel testing of 24 P. pyrifolia cultivars with unknown S-genotypes. In conclusion, the construction of c DNA microarrays has further improved the pear S-RNase detection platform. 展开更多
A Clinical Analysis of 293 FUO Patients, A Diagnostic Model Discriminating infectious Diseases from Non-infectious Diseases 预览

Methods The clinical data of patients with fever of unknown origin (FUO) hospitalized in Xiangya Hospital ... Objective A diagnostic model was established to discriminate infectious diseases from non-infectious diseases. Methods The clinical data of patients with fever of unknown origin (FUO) hospitalized in Xiangya Hospital Central South University, from January, 2006 to April, 2011 were retrospectively analyzed. Patients enrolled were divided into two groups. The ifrst group was used to develop a diagnostic model: independent variables were recorded and considered in a logistic regression analysis to identify infectious and non-infectious diseases (αin= 0.05, αout= 0.10). The second group was used to evaluate the diagnostic model and make ROC analysis. Results The diagnostic rate of 143 patients in the ifrst group was 87.4%, the diagnosis included infectious disease (52.4%), connective tissue diseases (16.8%), neoplastic disease (16.1%) and miscellaneous (2.1%). The diagnostic rate of 168 patients in the second group was 88.4%, and the diagnosis was similar to the ifrst group. Logistic regression analysis showed that decreased white blood cell count (WBC < 4.0×109/L), higher lactate dehydrogenase level (LDH > 320 U/L) and lymphadenectasis were independent risk factors associated with non-infectious diseases. The odds ratios were 14.74, 5.84 and 5.11 (P≤ 0.01) , respectively. In ROC analysis, the sensitivity and speciifcity of the positive predictive values was 62.1% and 89.1%, respectively, while that of negative predicting values were 75% and 81.7%, respectively (AUC = 0.76,P = 0.00). Conclusions The combination of WBC < 4.0×109/L, LDH > 320 U/L and lymphadenectasis may be useful in discriminating infectious diseases from non-infectious diseases in patients hospitalized as FUO. 展开更多

Objective:To explore the effect of emulsified isoflurane(EI)on apoptosis of anoxia-reoxygenation neonatal rat cardiomyocytea and relevant protein expression.Methods:Cardiac muscle anoxiareoxygenation damage model was established with culture in vitro neonatal rat cardiomyocytes.The cardiomyocytes were divided into control group,model group,fat emulsion group and EI group.The cardiomyocytes apoptosis rates and lactic dehydrogenase(LDH),superoxide dismutase(SOD)and malondialdehyde(MDA)index standardization were detected after relevant treatment The expression of apoptosis-related proteins Bel-2,Bax and Caspase-3 were detected with Western blot approach.Results:After hypoxia/reoxygenation(H/R)model was treated by EI,the cells apoptosis rate decreased and was dramatically below the fat emulsion group(P【0.05),Cardiomyocytes biochemical index detection presented that,compared with the control group that the LDH activity and MDA content dramatically increased(P【0.05),while the SOD activity notably decreased(P【0.05);compared with the H/R group,the SOD activity of the fat emulsion group and EI group increased(P【0.05);while the LDH activity and MDA content decreased(P【0.05).And the change of the EI group was more remarkable than the fat emulsion group(P【0.05).The Western blot analysis presented that,compared with the control group,the Bcl-2 protein expression of the other groups significantly decreased(P【0.05),the expressions of Bax protein and Caspase-3protein increased significantly(P【0.05);compared with H/R group,cardiomyocytes Bc1-2protein expression of EI group increased significantly(P【0.05),the expressions of Bax protein and Caspase-3 protein decreased significantly(P【0.05),and the change of EI group was more remarkable than the fat emulsion group(P【0.05).Conclusions:EI can inhabit the apoptosis of anoxia-reoxygenation damage model cardiomyocytes,and may he related to the up-regulation of expression of Bcl-2 and down-regulation of expression of Caspase-3 protein. 展开更多

Phytoremediation can be used as a sustainable technology for mine spoil remediation to remove heavy metals.This study investigated the concentration of 7 heavy metal contamination in soil and plant samples at an abandoned mine site.We found that,after vegetation remediation at the abandoned mine site,the reduction rates for 7 heavy metals were in the range of 4.2%-86%,where reduction rates over 50% were achieved for four heavy metals(Zn,Mn,Cd,Ni).Transfer coeffcients of the panicled goldenrain tree(Koelreuteria paniculata Laxm) and the common elaeocarpus(Elaeocarpus decipens) for Zn,Mn,Ni,and Co were more than 1.Enrichment coeffcients of both trees for Mn were higher than 1.Our results suggest that the panicled goldenrain tree and the common elaeocarpus tree may act as accumulators in remediation.Moreover,the woody vegetation remediation in abandoned mining areas play an important role in improving scenery besides removing heavy metal from contaminated soil. 展开更多

<正> Phytoremediation as a sustainable and inexpensive technology based on the removal of pollutants from the environmentby plants is becoming an increasingly important objective in plant research.In this study,... <正> Phytoremediation as a sustainable and inexpensive technology based on the removal of pollutants from the environmentby plants is becoming an increasingly important objective in plant research.In this study,biological cycles of five nutrientelements(N,P,K,Ca,and Mg)and eight heavy metal elements(Fe,Cu,Zn,Mn,Cd,Ni,Pb and Co)were examined in youngpaniculed goldraintree(Koelreuteria paniculata Laxm)and common elaeocarpus(Elaeocarpus decipens)mixed standsin an abandoned mining area.We found that after vegetation restoration in abandoned mining areas,the organic matterand concentrations of nutrient elements were significantly increased and the heavy metal elements were significantlydecreased,the annual retention,uptake and return were 75.0,115.4,and 40.3 kg/hm~2 for nutrient elements,and 1878.0,3231.0 and 1353.0 g/hm~2 for heavy metal elements,respectively,with the utilization coefficient,cycling coefficient andturnover rate of 0.92,0.35 and 0.32 for nutrient elements,and 1.24,0.42 and 1.92 for heavy metal elements,respectively.Our results suggested that the vegetation restoration in abandoned mining areas had significant effects in improvingenvironmental conditions,enhancing soil available nutrients,and ensuring human health. 展开更多

Expression of Pdx-1 in bone marrow mesenchymal stem cells promotes differentiation of islet-like cells in vitro 被引量:1

Bone marrow mesenchymal stem cells (BMSCs) have the ability of self-renewal and multi-directional differentiation. Recent reports showed that BMSCs could differentiate into endocrine cells of pancreas. However, the differentiation is not efficient enough to produce insulin-producing cells for the future therapeutic use. Pdx-1 is a crucial regulator for pancreatic development. Therefore we constructed a eukaryotic expression vector containing Pdx-1 to determine the effect of Pdx-1 expression on differentiation of BMSCs in vitro. The results showed that BMSCs could self-assemble to form functional pancreatic islet-like structures after differentiation in vitro. The proportion of insulin-producing cells differentiated from Pdx-1+BMSCs was 28.23%±2.56%, higher than that from BMSCs transfected with vacant vector and Pdx-1- BMSCs (7.23%±1.56% and 4.08%±2.69% respectively) by flow cytometry. Immunocytochemical examination also testified the expression of multiple β-cells-specific genes such as insulin, glucagons, somatostatin in differentiated BMSCs. The results also revealed that the expressions of genes mentioned above in Pdx-1+BMSCs were higher than that in Pdx-1-BMSCs, which was confirmed by Western blotting analysis and RT-PCR. Glucose-induced insulin secretion from Pdx-1+BMSCs in 5mmol/L and 25mmol/L glocuse was (56.61±4.82) μU/mLand (115.29±2.56) μU/mL respectively, which were much higher than those from Pdx-1-BMSCs((25.53±6.49) μU/mL and (53.26±7.56) μU/mL respectively). Grafted animals were able to maintain their body weight and survive for relatively longer periods of time than hyperglycemic sham-grafted controls,which demonstrated an overall beneficial effect of the grafted cells on the health of the animals. These findings thus suggested that exogenous expression of Pdx-1 should provide a promising approach for efficiently producing islet-like cells from BMSCs for the future therapeutic use in diabetic patients. 展开更多
