The Tribbles(TRIB) family of pseudokinase proteins has been shown to play key roles in cell cycle, metabolic diseases, chronic inflammatory disease, and cancer development. The Tribbles(TRIB) family of pseudokinase proteins has been shown to play key roles in cell cycle, metabolic diseases, chronic inflammatory disease, and cancer development. A better understanding of the mechanisms of TRIB pseudokinases could provide new insights for disease development and help promote TRIB proteins as novel therapeutic targets for drug discovery. At the 2 nd International Symposium on Tribbles and Diseases held on May 7–9, 2018 in Beijing, China, a group of leading Tribbles scientists reported their findings and ongoing studies about the effects of the different TRIB proteins in the areas of immunity, metabolism, fundamental cell biology and cancer. Here, we summarize important and insightful overviews from 4 keynote lectures, 13 plenary lectures and 8 short talks that took place during this meeting. These findings may offer new insights for the understanding of the roles of TRIB pseudokinases in the development of various diseases. 展开更多

Characterization of the salivary microbiome in healthy Thai children

Objective: To investigate the composition of the salivary microbiome of 50 healthy Thai children. Methods: A total 76 provinces in Thailand are grouped into 5 geographical clusters based on unique economics, foods and lifestyles. Geographical locations and the results of an oral assessment were also considered. Genomic DNA was extracted from stimulated saliva samples. Subsequently, amplicon libraries were prepared by 16 S Metagenomic Sequencing Library Preparation. The amplicons were sequenced using an Illumina Miseq platform followed by bioinformatics and statistical analyses.Results: The correlation between oral hygiene status and caries history varied from r~2=0.887 to r~2=0.999 in the geographical groups, suggesting oral hygiene status a strong association between caries history. Twenty taxonomic groups were found in all subjects and constituted 93.6%-96.5% of the microbiome. Of these, genus Veillonella and Prevotella showed significant differences in their proportions between the geographical groups(P<0.05). Furthermore, the proportion of Veillonella parvula, as well as Rothia aeria and Rothia dentocariosa tended to increase with worse oral hygiene status, which was also related to higher dental caries history.Conclusions: The differences in the salivary microbiome as related to geographic regions suggest that environmental factors, which may include dietary habits, could influence the predominant bacteria found in the mouth of Thai children, especially the genus Veillonella and Prevotella. The ratio of Veillonella parvula, Rothia aeria and Rothia dentocariosa may be indicators of worse oral hygiene status and future caries in this population. 展开更多

We previously est... Transplantation of oligodendrocyte progenitor cells (OPCs) is a promising way for treating demyelinating diseases. However, generation of scalable and autologous sources of OPCs has proven difficult. We previously established a chemical condition M9 that could specifically initiate neural program in mouse embryonic fibroblasts. Here we found that M9 could induce the formation of colonies that undergo mesenchymal-to-epithelial transition at the early stage of reprogramming. These colonies may represent unstable and neural lineage-restricted intermediates that have not established a neural stem cell identity. By modulating the culture signaling recapitulating the principle of OPC development, these intermediate cells could be reprogrammed towards OPC fate. The chemical-induced OPC-like cells (ciOPLCs) resemble primary neural stem cell-derived OPCs in terms of their morphology, gene expression, and the ability of self-renewal. Upon differentiation, ciOPLCs could produce functional oligodendrocytes and myelinate the neuron axons in vitro, validating their OPC identity molecularly and functionally. Therefore, our study provides a non-integrating approach to OPC reprogramming that may ultimately provide an avenue to patient-specific cell-based or in situ regenerative therapy. 展开更多

Impact of high-fat diet and vitamin D3 supplementation on aortic stenosis establishment in waved-2 epidermal growth factor receptor mutant mice

Objective: The use of an imal models of aortic stenosis (AS) remains essential to further elucidate its pathophysiology and to evaluate new therapeutic strategies. The waved-2 mouse AS mode! has been proposed;data have indicated that while aortic regurgitation (AR) is effectively induced, development of AS is rare. We aimed to evaluate the effect of high-fat diet (HFD) and vitamin D3 supplementation in this model. Methods: HFD and subcutaneous vitamin D3 injections were initiated at the age of 6 weeks until the age of 6 (n = 16, 6-month treatme nt group) and 9 (n = 11, 9-m onth treatment group) mon ths. Twelve waved-2 mice without supplementation were used as control. Echocardiography was performed at 3, 6 and 9 months. Blood serum analysis (calcium, 1,25(OH)2D3 and cholesterol), histology and immunohistochemistry (CD-31, CD-68 and osteopontin) were evaluated at the end of the experiment (6 or 9 months). Results: Total cholesterol and 1,25(OH)2D3 were significantly increased relative to the control group. HFD and vitamin D3 supplementation did result in improvements to the model, since AS was only detected in 6 (15.3%) mice (2 in the 3 groups) and AR was developed in the remaining animals. Echocardiographic parameters, fibrosis, thickness, inflammation and valvular calcification, were not significantly different between the 6-month treatment and control groups. Similar results were also observed in the 9-month treatment group. Conclusion: These results suggest that HFD and vitamin D3 supplementation have no effect in the waved-2 mouse model. This model essentially mimics AR and rarely AS. Further studies are needed to find a reliable animal model of AS. 展开更多

Successful fracture healing requires the simultaneous regeneration of both the bone and vasculature;mesenchymal stem cells (MSCs) are directed to replace the bone tissue, while endothelial progenitor cells (EPCs) form the new vasculature that supplies blood to the fracture site. In the elderly, the healing process is slowed, partly due to decreased regenerative function of these stem and progenitor cells. MSCs from older individuals are impaired with regard to cell number, proliferative capacity, ability to migrate, and osteochondrogenic differentiation potential. The proliferation, migration and function of EPCs are also compromised with advanced age. Although the reasons for cellular dysfunction with age are complex and multidimensional, reduced expression of growth factors, accumulation of oxidative damage from reactive oxygen species, and altered signaling of the Sirtuin-1 pathway are contributing factors to aging at the cellular level of both MSCs and EPCs. Because of these geriatric-specific issues, effective treatment for fracture repair may require new therapeutic techniques to restore cellular function. Some suggested directions for potential treatments include cellular therapies, pharmacological agents, treatments targeting age-related molecular mechanisms, and physical therapeutics. Advanced age is the primary risk factor for a fracture, due to the low bone mass and inferior bone quality associated with aging;a better understanding of the dysfunctional behavior of the aging cell will provide a foundation for new treatments to decrease healing time and reduce the development of complications during the extended recovery from fracture healing in the elderly. 展开更多

To examine the effects of Populus tomentiglandulosa(PT) extract on the expressions of antioxidant enzymes and neurotrophic factors in the cornu ammonis 1(CA1) region of the hippocampus at 5 min after inducing transient global cerebral ischemia(TGCI) in gerbils, TGCI was induced by occlusion of common carotid arteries for 5 min. Before ischemic surgery, 200 mg·kg–1 PT extract was orally administrated once daily for 7 d. We performed neuronal nuclear antigen immunohistochemistry and Fluoro-Jade B staining. Furthermore, we determined in situ production of superoxide anion radical, expression levels of SOD1 and SOD2 as antioxidant enzymes and brain-derived neurotrophic factor(BDNF) and insulin-like growth factor I(IGF-I) as neurotrophic factors. Pretreatment with 200 mg·kg–1 PT extract prevented neuronal death(loss). Furthermore, pretreatment with 200 mg·kg–1 PT extract significantly inhibited the production of superoxide anion radical, increased expressions of SODs and maintained expressions of BDNF and IGF-I. Such increased expressions of SODs were maintained in the neurons after IRI. In summary, pretreated PT extract can significantly increase levels of SODs and protect the neurons against TGCI, suggesting that PT can be a useful natural agent to protect against TGCI. 展开更多

Gastroduodenal disease(GDD)was initially thought to be uncommon in Africa.Amongst others,lack of access to optimal health infrastructure and suspicion of conventional medicine resulted in the reported prevalence of GDD being significantly lower than that in other areas of the world.Following the increasing availability of flexible upper gastro-intestinal endoscopy,it has now become apparent that GDD,especially peptic ulcer disease(PUD),is prevalent across the continent of Africa.Recognised risk factors for gastric cancer(GCA)include Helicobater pylori(H.pylori),diet,Epstein-Barr virus infection and industrial chemical exposure,while those for PUD are H.pylori,non-steroidal antiinflammatory drug(NSAID)-use,smoking and alcohol consumption.Of these,H.pylori is generally accepted to be causally related to the development of atrophic gastritis(AG),intestinal metaplasia(IM),PUD and distal GCA.Here,we perform a systematic review of the patterns of GDD across Africa obtained with endoscopy,and complement the analysis with new data obtained on premalignant gastric his-topathological lesions in Accra,Ghana which was compared with previous data from Maputo,Mozambique.As there is a general lack of structured cohort studies in Africa,we also considered endoscopy-based hospital or tertiary centre studies of symptomatic individuals.In Africa,there is considerable heterogeneity in the prevalence of PUD with no clear geographical patterns.Furthermore,there are differences in PUD within-country despite universally endemic H.pylori infection.PUD is not uncommon in Africa.Most of the African tertiary-centre studies had higher prevalence of PUD when compared with similar studies in western countries.An additional intriguing observation is a recent,ongoing decline in PUD in some African countries where H.pylori infection is still high.One possible reason for the high,sustained prevalence of PUD may be the significant use of NSAIDs in local or over-the-counter preparations.The prevalence of AG and IM,were similar or modestly higher ove 展开更多

This study aimed to investigate the effects of agnuside(AGN),a phytoestrogen in Vitex agnus-castus,on cellular and metabolic alterations induced by ovariectomy in rats.Ovariectomized(OVX)Wistar rats were used as an animal model of postmenopausal metabolic syndrome.The visceral and subcutaneous adipocyte sizes,the peripheral insulin sensitivity and the occurrence of nonalcoholic fatty liver disease(NAFLD)and liver oxidative damage were evaluated.The results were compared with those obtained from control rats(sham-operated)and OVX rats treated daily with AGN.Obese OVX rats exhibited visceral adipocyte hypertrophy and developed glucose intolerance,hyperinsulinemia,NAFLD and dyslipidemia.AGN exerted beneficial effects on several metabolic parameters in OVX rats,including the improvement in glucose intolerance,hyperinsulinemia and NAFLD.Besides,AGN promoted a general improvement in the liver redox state.Although the reduction of the mesenteric adipocyte size is very likely to be an important mode of action of AGN,some actions of AGN could also involve the interaction with estrogen receptors. 展开更多

The plant circadian clock regulates the rhythms of plant metabolism.Many herbivore-induced plant volatiles(HIPVs)fluctuate,diurnally,but the role of the circadian clock in the emission of HIPVs and their ecological consequences remains largely unknown.Here,we show that the timing of herbivore attack can alter the outcome of tri-trophic interactions,and this is mediated by the circadian clock,under both field and glasshouse conditions.Although most HIPV emissions did not have a circadian rhythm,the circadian clock modulated HIPV emissions in a time-dependent manner.HIPVs mediate tri-trophic interactions,and the circadian clock may affect these interactions by modulating HIPV emission in nature. 展开更多

Objective: To determin the chemical compounds of Mentha suaveolens(M. suaveolens) and Pinus halepensis(P. halepensis) essential oils(Eos) and evaluate their antioxidant and antibacterial activities. Methods: The chemical composition of P. halepensis and M. suaveolens EOs was determined by GC-MS analysis. The antioxidant activity was evaluated using DPPH,ABTS and FRAP assays. The antibacterial effect was tested against 6 bacterial strains using the well diffusion method and micro-dilution assay. Results: The major components of P. halepensis EOs were β-caryophyllene(28.04%),myrcene(23.81%) and α-pinene(12.02%). However,piperitenone oxid(56.28%),piperitenone(11.64%) and pulegone(6.16%) were the major components of M. suaveolens EOs. M. suaveolens EOs showed remarkable antioxidant activities compared with P. halepensis EOs,showing antioxidant capacity values of IC50=(64.76±2.24) μg/mL,IC50=(82.73±3.34) μg/mL,and IC50=(93.35±4.45) μg/mL,revealed by DPPH,ABTS and FRAP assays,respectively. However,P. halepensis EOs showed interesting antibacterial effects against all bacterial strains. The most sensible strains to P. halepensis EOs were Staphylococcus aureus [(34.00±0.50) mm],Listeria monocytogenes [(31.00±1.50)] mm and Proteus mirabilis ((29.00±2.25)mm)Furthermore,the lowest minimal inhibitory concentration(MIC) and minimal bactericidal concentration(MBC) values were revealed by P. halepensis EOs against Staphylococcus aureus [MIC=MBC=0.125%(v/v)] and Listeria monocytogenes (MIC=MBC=0.25%(v/v))Conclusions: P. halepensis and M. suaveolens EOs contain bioactive compounds that could have potential applications against bacterial infections and oxidative stress related diseases as well as for food conservation. Group members may mediate conflict through submissive and affiliative behaviors, which can reduce aggression, stabilize dominance hierarchies, and foster group cohesion. The frequency and resolution of within-group conflict may vary with the presenee of neighboring groups. Neighbors can threaten the territory or resources of the whole group, promoting behaviors that foster within-group cohesion. However, neighbors may also foster conflict of interests among group members: opportunities for subordinate dispersal may alter conflict among dominants and subordinates while opportunities for extrapair reproduction may increase conflict between mates. To understand how neighbors mediate within-group conflict in the cooperatively breeding fish Neolamprologus pulcher, we measured behavioral dynamics and social network structure in isolated groups, groups recently exposed to neighbors, and groups with established neighbors. Aggression and submission between the dominant male and female pair were high in isolated groups, but dominant aggression was directly primarily at subordinates when groups had neighbors. This suggests that neighbors attenuate conflict between mates and foster conflict between dominants and subordinates. Further, aggression and submission between similarly sized group members were most frequent when groups had neighbors, suggesting that neighbors induce rank-related conflict. We found relatively little change in within-group affiliative networks across treatments, suggesting that the presence of neighbors does not alter behaviors associated with promoting group cohesion. Collectively, these results provide some of the first empirical insights into the extent to which intragroup behavioral networks are mediated by intergroup interactions and the broader social context. 展开更多

Plant organelles, with various functions,migrate along actin filaments and contact other types of organelles, leading to physical interactions at a specific site called the membrane contact site. Recent studies have revealed the importance of physical interactions in maintaining efficient metabolite flow between organelles.In this review, we first summarize peroxisome function under different environmental conditions and growth stages to understand organelle interactions. We then discuss current knowledge regarding the interactions between peroxisome and other organelles, i.e., the oil bodies, chloroplast, and mitochondria from the perspective of metabolic and physiological regulation, with reference to various organelle interactions and techniques for estimating organelle interactions occurring in plant cells. 展开更多

Can house sparrows recognize familiar or kin-related individuals by scent?

As a consequence, a handful of avian species have been discovered to use olfaction in different contexts. Nevertheless, we still have a very limited knowledge about the use of odor cues in avian social life, particularly, in the case of songbirds. Here, we investigate if female house sparrows Passer domesticus show any preference for the odor of kin and nonkin conspecifics and we also test a possible role of familiarity based on male see nt in female choice. We performed the experiment with captive birds twice, duri ng the non breeding and breedi ng seas ons. Our results show that female house sparrows strongly avoided the odor of unrelated familiar (UF) males, both in the breeding and nonbreeding seasons. Our results suggest recognition for olfactory stimuli related to familiarity and kin ship. We suggest that avoidance for UF males is associated with previous experie nee in this species. Also, we provided further evidenee to the use of olfaction in passerine species by using a new experimental setup. 展开更多

Mitophagy is activated by a number of stimuli,including hypoxia,energy stress,and increased oxidative phosphorylation activity.Mitophagy is associated with oxidative stress conditions and central neurodegenerative diseases.Proper regulation of mitophagy is crucial for maintaining homeostasis;conversely,inadequate removal of mitochondria through mitophagy leads to the generation of oxidative species,including reactive oxygen species and reactive nitrogen species,resulting in various neurodegenerative diseases,such as Alzheimer’s disease,Parkinson’s disease,Huntington’s disease,and amyotrophic lateral sclerosis.These diseases are most prevalent in older adults whose bodies fail to maintain proper mitophagic functions to combat oxidative species.As mitophagy is essential for normal body function,by targeting mitophagic pathways we can improve these disease conditions.The search for effective remedies to treat these disease conditions is an ongoing process,which is why more studies are needed.Additionally,more relevant studies could help establish therapeutic conditions,which are currently in high demand.In this review,we discuss how mitophagy plays a significant role in homeostasis and how its dysregulation causes neurodegeneration.We also discuss how combating oxidative species and targeting mitophagy can help treat these neurodegenerative diseases. 展开更多

Objective:An extract of Costus speciosus(CSE),a herb widely used in folk medicine,was evaluated for its antioxidant,antihyperlipidemic and ameliorating effects on histopathological changes in atherogenic rabbits.Methods:Twenty-four male rabbits(Oryctolagus cuniculus)were divided into 4 groups.Three groups were fed a diet containing 3%saturated fat and 1.3%cholesterol for 40 d.One of these was sacrificed on the 40th day and was called the pathogenic(P)group;the other two groups received treatment for another 30 d as follows:one received 0.8 g/(kg·d)of CSE and the other was given 0.01 g/(kg·d)of simvastatin.The normal group was sacrificed on the 70th day and used as a control.Results:CSE showed radical-scavenging ability.Administration of CSE for a 30-day period resulted in a significant decrease in total cholesterol,triacylglycerol,low-density lipoprotein and aspartate aminotransferase compared to the P group,while levels of hemoglobin,packed corpuscular volume and red blood cells were elevated.With respect to studies performed on the heart,a decrease in malondialdehydeand an increase in reduced glutathione were noted.Total protein increased in the liver,heart and aorta after treatment with CSE and also a marked improvement in histopathological parameters was demonstrated.Conclusion:The present findings indicate that the C.speciosus rhizome possesses antiatherogenic and antioxidant properties which may provide protective effects against oxidative stress in atherosclerotic rabbits. 展开更多

The recognition that neurogenesis does not stop with adolescence has spun off research towards the reduction of brain disorders by enhancing brain regeneration. Adult neurogenesis is one of the tougher problems of developmental biology as it requires the generation of complex intracellular and pericellular anatomies, amidst the danger of neuroinflammation. We here review how a multitude of regulatory pathways optimized for early neurogenesis has to be revamped into a new choreography of time dependencies. Distinct pathways need to be regulated, ranging from neural growth factor induced differentiation to mitochondrial bioenergetics, reactive oxygen metabolism, and apoptosis. Requiring much Gibbs energy consumption, brain depends on aerobic energy metabolism, hence on mitochondrial activity. Mitochondrial fission and fusion, movement and perhaps even mitoptosis, thereby come into play. All these network processes are interlinked and involve a plethora of molecules. We recommend a deep thinking approach to adult neurobiology. 展开更多
