目的 了解红霉素对肺纤维化的防护作用。方法 采用大鼠油酸尾静脉注射制作急性肺损伤后肺纤维化模型 ,观察红霉素对损伤肺组织中中性粒细胞聚集及氧自由基的干预作用。结果 血清检测 :油酸注射后第 12天和第 2 4天的MDA明显升高 ,而SOD明显降低 ,MDA和SOD呈负相关 (r =- 0 .91) ;红霉素干预后GSH明显降低 ;注射油酸组GSH -Px水平最高 ,红霉素干预后GSH -PX水平最低。组织匀浆检测 :MDA变化同血清 ;SOD、GSH组间无差异 ;油酸注射后第 12天和第 2 4天的GSH -Px水平明显低于红霉素干预组。镜下观察 :注射油酸组中性粒细胞浸润和纤维化程度明显重于红霉素干预组。结论 红霉素可通过抑制中性粒细胞而减少氧自由基的产生 ,减轻纤维化程度 ,对油酸所致急性肺损伤后的肺纤维化有一定防护作用
Objective: To understand the prophylaxis effect of erythromycin on fibrosis of lung .Methods: Model of fibrosis of lung with injection by oil-acid to rat, observed the effect of erythromycin to neutrophil and oxygen-free radical. Result: Serum test: MDA increased obviously after injection oil-acid 12 and 24days, but SOD decreased obviously, MDA and SOD showed negative correlation(-0.91). GSH decreased after use of Erythromycin; GSH-PX was lowest in using oil-acid and highest in using Erythromycin. Tissue liquid test: The change of MDA was similar to serum; SOD and GSH had no difference among groups. 12 and 24 days after of injection oil-acid, GSH-PX was lower obviously lower than group of Erythromycin. Micrography: Congestion and neutrophil infiltration in the group of oil-acid injection was severer than those in group using erythromycin.Conclusion: Erythromycin can reduce the production of oxygen-free radical by repressing neutrophil, subdue inflammation and exudation of alveolus, mitigate chronic prompting of injury factor and prevent fibrosis of lung . Erythromycin has prophylaxis effect on fibrosis of lung with oil-acid.
