槲皮素对百草枯致大鼠急性肺损伤治疗作用的实验研究 认领 被引量:3

Therapeutic effects of quercetin against paraquat-induced acute lung injury in rats

摘要 目的 探讨槲皮素对百草枯所致急性肺损伤的治疗作用。方法 雄性SD大鼠45只随机分为对照组(5只)、百草枯组(20只)和槲皮素组(20只),后两组均再分为8,24,48和72 h四个小组(每组5只)。百草枯组和槲皮素组大鼠腹腔注射百草枯(50 mg/kg),槲皮素组在百草枯注射后3 h再腹腔注射槲皮素(100 mg/kg)。分别测定百草枯中毒后8,24,48,72 h大鼠血浆和支气管肺泡灌洗液(BALF)中丙二醛(MDA)含量、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)的活力,并观察肺组织结构改变,测定肺组织湿/干重比(W/D),采用免疫组化方法测定肺组织核转录因子κB(NF-κB)p65的表达。结果 与对照组比较,百草枯组血浆及BALF中MDA含量均显著升高;血浆中SOD活力呈下降趋势,24 h后明显下降;GSH-Px活力呈增加趋势,在48 h后明显升高;BALF中SOD、GSH-Px活力均下降,但48 h后SOD活力显著升高。槲皮素组与百草枯组比较,血浆和BALF中MDA含量下降,GSH-Px和SOD活力升高。百草枯组肺泡壁充血、增厚,炎症细胞浸润,并有局灶性出血;槲皮素组肺泡壁充血、出血均有减轻。槲皮素组肺组织NF-κB p65的表达及W/D值较百草枯组明显下降。结论 槲皮素对百草枯诱导的急性肺损伤具有治疗作用。 Objective To study the therapeutic effects of quercetin against paraquat-induced acute lung injury. Methods Forty-five male SD rats were randomly divided into three groups, ie, a control group( n = 5 ), a paraquat group( n = 20), and a quercetin group( n = 20). The rats in the paraquat group and quercetin group were all injected intraperitoneally with paraquat(50 mg/kg) to induce lung injury, then the quercetin group was injected quercetin( 100 mg/kg) 3 h later. Five rots in each group were sacrificed at 8,24,48,72 h respectively, The levels of malondialdehyde(MDA), superoxide dismutade(SOD) and glutathione peroxidase(GSH-Px) both in plasma and bronchoalveolar lavage fluid(BALF) were detected. Expression of NF-κB p65 in lung was measured by immunohistochemistry staining, and lang wet/dry weight ratio(W/D) were recorded. Results Comparing the paraquat group with the control group, MDA level increased significantly both in plasma and BALF, SOD activitie decreased both in plasma and BALF but increased significantly in BALF 48 h later, and GSH-Px increased siganificently in plasma 48 h later and decreased in BALF. After the quercetin treatment, MDA decreased, SOD and GSH-Px activities increased significantly both in plasma and BALF. The congestion and hemorrhage of alveoli got improvement.The expression of nuclear factor kappa B(NF-κB) p65 in lungs and W/D were decreased as well in the quercetin group( P 〈 0.05). Conclusion Quercetin has therapeutic effects on paraquat-induced acute lung injury in rats.
