吉林梅花鹿肌肉发育关联基因MyoG的鉴定及生物信息学分析 认领

Identification and Bioinformatics Analysis of MyoG Gene Related to Muscle Development in Jilin Sika Deer(Cervus nippon)

摘要 为探明吉林梅花鹿MyoG基因的分子结构特征与生理功能,本研究通过提取吉林梅花鹿血液基因组DNA,利用PCR技术分段扩增、Sanger测序并拼接获取MyoG基因,对MyoG基因核苷酸及编码氨基酸序列的分子结构特征进行了生物信息学分析。结果表明,吉林梅花鹿MyoG基因序列长3162 bp(GenBank登录号:HZ56689),含有3个外显子、2个内含子及部分5 UTR和3 UTR,编码区(CDS)为684 bp,共编码227个氨基酸,与NCBI中甘肃马鹿、水牛及山羊该基因相似性分别为99%、94%和92%;通过分析推导氨基酸序列结构发现,1~89位氨基酸为MYOD家族特有的碱性结构域,85~136位为螺旋-环-螺旋DNA结合结构域,含有21个激酶磷酸化的位点;吉林梅花鹿与甘肃马鹿该基因CDS区存在6个突变位点,导致3个氨基酸位点发生了变异;序列相似性及分子系统进化分析显示,吉林梅花鹿MyoG基因与反刍动物山羊、绵羊及水牛的亲缘关系较近。吉林梅花鹿MyoG基因的克隆及序列结构特征的生物信息学预测分析为解析鹿科动物肌肉生长发育的分子调控机制奠定了理论基础。 To explore the structure and physiological function of MyoG gene in Jilin sika deer,the MyoG gene of Jilin sika deer was amplified through PCR technology using extracted genomic DNA from blood samples as template.Then the structural characteristics of nucleotide and amino acid sequence were analyzed by bioinformatics tools.The results showed that the length of Jilin sika deer MyoG gene was 3162 bp(GenBank accession number:HZ56689)and consisted 3 exons,2 introns and partial of 5 UTR(381 bp)and 3 UTR(690 bp)untranslated region,which contained an open reading frame of 684 bp and encoded 227 amino acids.The sequence similarity with Gansu wapiti,buffalo and goat deposited in GenBank database was 99%,94%and 92%,respectively.Amino acid sequence structure prediction showed that 1-89 amino acids were the basic domain of MYOD family,and 85-136 amino acids were the helix-loop-helix DNA binding domain,containing21 sites that could be phosphorylated by kinase.There were six mutation sites between Jilin sika deer and Gansu wapiti in CDS,resulting in three amino acid mutations.Sequence similarity and molecular phylogenetic analysis showed that Jilin sika deer MyoG gene was closely related to goat,sheep and buffalo,which belongs to ruminant.Bioinformatics prediction of Jilin sika deer MyoG gene would lay theoretical foundation for further study on molecular regulatory mechanism of muscle growth and development in the Cervidae.
